玉树地震受伤患者伤口感染梭状芽胞杆菌培养鉴定分析

摘要
目的了解玉树地震受伤患者伤口感染梭状芽胞杆菌情况,为临床诊治及突发公共卫生灾害细菌的感染救治提供参考依据。方法 3例伤口感染的地震受伤患者入院时立即采集伤口分泌物进行细菌厌氧和需氧培养,并采用ATB自动微生物分析仪进行细菌鉴定;鉴定出的梭状芽胞杆菌菌株经PCR扩增细菌16S rRNA基因,DNA测序分析,进一步确定细菌种类。结果 3份标本经自动微生物分析仪鉴定出1株产气荚膜梭菌、4株疑似梭状芽胞杆菌;PCR扩增显示5株菌均出现目的大小条带;测序证实5株菌有1株为产气荚膜梭菌,另4株为需氧芽胞杆菌属细菌。结论自动微生物分析仪鉴定和细菌16S rRNA基因测序分析联合应用可快速、准确为临床提供诊治和预防控制信息。
OBJECTIVE To investigate the status of wounds infection of Clostridium from Yushu earthquake survivors accepted by our hospital,and to provide diagnostic and therapeutic information of bacteria for clinic and public health disaster.METHODS Secretion was collected from wounds of three earthquake victims as soon as hospitalized and performed anaerobic and aerobic cultivation immediately.Bacteria were identified by ATB automatic microbiotic analyzer.16S rRNA gene was amplified by PCR and DNA sequencing was applied to further confirm bacteria species.RESULTS Bacteria were characterized by ATB automatic microbiotic analyzer,including one Clostridium perfringens and four suspectable Clostridia.5 isolates showed PCR positive results.One isolate was identified as Clostridium perfringens and the others were identified as Bacillus by DNA sequencing.CONCLUSION Associated use of ATB automatic microbiotic analyzer and 16S rRNA gene sequencing can provide diagnostic and therapeutic information to clinic and public health disaster quickly and exactly.
