芦荟多糖对大鼠局灶性脑缺血损伤的保护作用研究

摘要
目的:观察芦荟多糖对局灶性脑缺血大鼠的神经保护作用,并进一步探讨其作用机制。
     方法:采用线栓法阻断大鼠一侧大脑中动脉(MCAO),制备局灶性脑缺血模型,观察芦荟多糖对MCAO模型大鼠脑缺血后24h神经功能缺损导致的行为学变化,通过HE染色及透射电镜观察MCAO模型大鼠脑组织病理形态学变化,此外还通过流式细胞仪检测芦荟多糖对MCAO模型大鼠脑缺血后24h大脑皮质细胞凋亡率的影响,并以免疫组化法检测芦荟多糖对其大脑皮质Caspase-3、COX-2、NF-κBp65、ICAM-1蛋白表达的影响,以此探讨芦荟多糖对脑缺血的神经保护作用及其作用机制。
     结果:芦荟多糖能明显改善MCAO模型大鼠神经功能缺损的症状和脑缺血组织的病理形态学变化,降低缺血脑组织的凋亡率,抑制NF-κBp65、ICAM-1、Cox-2、Caspase-3蛋白表达。
     结论:芦荟多糖对大鼠局灶性脑缺血具有保护作用,其机理可能与芦荟多糖抗炎和抑制神经元细胞凋亡有关。
Objective:To study and explore the neuroprotective effects and thefurther mechanism of polysaccharide of Aloe barbadensis(PAB) oncerebral ischemia in rats.
     Methods:In order to observe the neuroprotective effect of PAB oncerebral ischemia,the rats with incomplete cerebral ischemia were inducedwith fishing lines in the middle cerebral artery(MCA) and then wereexecuted at 24h after cerebral ischemia.It was observed the effect of PABon the changes of ethology and pathomorphology due to nerve injury ofrats with MCAO before and after treatments.HE staining was used toobserve the changes of histomorphology.Electronic microscopy was usedto study the ultrastructure of areas.It was detected the effects of PAB onprotein expression ofcysteinyaspartate specificprotease-3(Caspase-3),Cyclooxygenase-2(COX-2),Nuclear Factor Kappa B p65(NF-κB p65)and intercellular adhesion molecule-1(ICAM-1) with immunohistochemicalmethod.In addition,we observe the rate of cell apoptosis with flowcytometry.
     Results:PAB could make obviously pathomorphology of cerebralischemia better,improve change of ethology,decrease the rate of cellapoptosis of ischemic cerebral tissue,inhibit the expression of cysteinyaspartate specific protease-3(Caspase-3),Cyclooxygenase-2(COX-2),Nuclear Factor Kappa B p65(NF-κB p65) and intercellular adhesionmolecule-1(ICAM-1) protein in cerebral cortex to protect the nerves aftercerebral ischemic injury of rats.
     Conclusion:PAB had protecting effect on cerebral ischemia,themechanism of PAB against cerebral ischemia is related toanti-inflammatory action and inhibiting cell apoptosis.
