基于质谱技术的中药药效物质基础和作用机制研究

摘要
由于中药成分的复杂性、体内处置过程的多样性以及作用过程的整体性,给中药药效物质基础及作用机制研究带来了极大的困难和挑战。本课题组充分发挥现代质谱及联用技术快速、灵敏及特异性强的优势,建立了系统的质谱分析新方法和创新研究平台,并将其应用于中药药效物质基础及作用机制研究中,开展了较为深入的研究工作,取得了一系列重要成果:从离子内能角度阐明各类中药成分的质谱特征规律,成功建立了皂苷、黄酮和寡糖等多类成分简便、灵敏、快速的质谱分析方法;基于中药成分与生物靶分子的相互作用,选择糖尿病、肿瘤等疾病相关靶分子,建立了超滤质谱、强度衰减质谱、流动注射亲和质谱等中药活性成分筛选方法;建立了微透析质谱在线联用方法,并应用于中药体内过程及作用机制研究;建立了基于质谱技术的中药代谢组学研究方法,从中药作用的整体性角度阐明了中药复方乌头汤治疗佐剂性关节炎的药效物质基础及作用机制。可以说,质谱及其相关联用技术作为探究中医药科学内涵的利器,极大的推动了中药创新性研究的进程。
A series of innovative mass spectrometry and its hyphenated methods were established by our group,and these methods were used to study the pharmacodynamic material basis and function mechanism of Traditional Chinese Medicine.The achievements are as follows:(1) \The mass spectrometry method for rapid structural analysis of saponins,flavonoids and oligosaccharides was successfully established.(2) Ultrafiltration mass spectrometry method,intensity-fading mass spectrometry method and flow injection affinity mass spectrometry method were developed for screening active ingredients from Traditional Chinese Medicine.(3) Online microdialysis coupled with liquid chromatography mass spectrometry(MD-LC/MS) was applied to investigating the in vivo process and function mechanism of active ingredients.(4) The pharmacodynamic material basis and function mechanism of Wu-tou formula for treating rheumatoid arthritis were integrated clarified based on the metabonomic method using mass spectrometry.
