浙江地方柚类种质资源遗传关系的SSR标记研究

浙江省是我国柚类遗传多样性的主要分布区之一,本研究搜集本省18份地方柚类种质资源利用24对柑橘SSR引物进行遗传多样性分析并构建种质鉴定图,旨在为地方资源的搜集保护和合理利用提供参考。24对引物共检测到92个等位变异位点,变异范围在1-7之间,平均每对SSR引物可检测到3.8个等位位点;24个SSR位点的观察杂合度、期望杂合度和多态性信息含量分别为0.49、0.51和0.45;18份柚类种质资源两两间的遗传相似性系数介于0.55-0.94之间,平均为0.71,表明浙江地方柚类种质资源具有丰富的遗传多样性,遗传背景较复杂。UPGMA聚类分析将供试材料分成A1-A3、B1、C1和D1共6个组群,其中A1组包括四季柚、佛香柚和麻步文旦等7份果肉颜色为黄色或粉红色的种质,A2组包括永嘉红心柚2号、青田红心柚和木兰柚3份果肉颜色为浅红色的种质,A3组包括处红柚、永嘉红心柚1号、古磉柚和官南红柚4份果肉颜色为红色的种质,官南土柚和文成红心柚自成B1和C1组,常山胡柚和槾文柑2个杂种柚类型聚为D1组。除少数个别材料外,浙江地方柚类种质资源有按果肉颜色优先聚类的趋势,但是黄色和粉红色的种质资源间并未得到明显的区分,说明SSR分子聚类结果与果实表型基本吻合。另外,利用AG14、CAG01、CMS-16和CMS-26 4对SSR引物可快速准确地将18份供试种质鉴别开来,以此构建了浙江省地方柚类资源的种质鉴定图,可应用于种苗真实性鉴定。
It is one of the main distribution area of pummelo genetic diversity in Zhejiang province.In this study,twenty-four simple sequence repeat(SSR) primers developed in Citrus were used to detect the genetic diversity of 18 local pummelo germplasms collected in Zhejiang province,and to construct germplasm characterization map,which would contribute to the local resources protection and subsequent rational utilization.A total of 92 alleles were detected among twenty-four SSR loci,ranged from one to seven and with an average of 3.8 alleles per locus.The mean observed and expected heterozygosity values were respectively 0.49 and 0.51,and the average polymorphism information content of 24 SSR loci was0.45.The genetic similarity coefficients between genotypes ranged from 0.55 to 0.94 and with an average of 0.71.The above results showed a rich genetic polymorphism and complicated genetic background among Zhejiang pummelo germplasm resources.The UPGMA dedrogram based on the genetic similarity coefficients grouped 18 pummelo accessions to six major groups at the coefficient of 0.71 as follows:Sijiyou,Foxiangyou,Mabu-wendan and other seven yellow or pink pulp colour cultivars were clustered to A1 group;the three pale red cultivars including Yongjia-hongxinyou 2,Qingtian-hongxinyou and Mulanyou were clustered to A2 group;the four red cultivars including Chuhongyou,Yongjia-hongxinyou 1,Gusangyou and Guannan-hongyou were clustered to A3 group;Guannan-tuyou and Wencheng-hongxinyou was separately clustered to B1 and C1;and the two hybrids from the cross of pummelo and other citrus were clustered to D1 group.The results showed that except for a few germplasms most local pummelo cultivars were clustered preferentially related to their pulp colours,however,the germplasms between yellow and pink colours were not totally distingushed,which indicated the SSR molecular dendrogram approximately agreed with the morphological characters.Moreover,four SSR primers AG14,CAG01,CMS-16 and CMS-26 could rapidly and accurately separate all the tested materials apart,and accordingly a germplasm characterization map of Zhejiang province was constructed,which could be applied to seedling authenticity characterization.
