山楂葛根颗粒对冠脉结扎大鼠心肌缺血保护作用研究

摘要
目的:通过山楂葛根颗粒对冠脉结扎大鼠心肌缺血的影响作用研究,探讨其对大鼠心肌细胞凋亡影响,进而探讨相关机制。方法:将60只SD大鼠雄性均分为6组,每组10只,分别为假手术组、模型组、山楂葛根颗粒低剂量组、山楂葛根颗粒中剂量组、山楂葛根颗粒高剂量组、硝酸异山梨酯组。构建大鼠心肌缺血再灌注损伤模型-结扎左冠状动脉前降支(LAD)30 min再灌注2 h。大鼠腹主动脉采血用免疫分析法检测血清肌钙蛋白,大鼠心肌组织呈现状态采用苏木素伊红染色法(HE)进行观察,心肌凋亡相关蛋白Caspase-3的表达和AIF的表达采用免疫组化法检测。结果:与模型组相比较,山楂葛根颗粒中剂量、高剂量组和硝酸异山梨酯组血清肌钙蛋白呈降低趋势,HE染色心脏病理分级变化程度显著,心肌细胞Caspase-3表达显著性降低,而AIF表达减弱不显著无统计学意义。结论:山楂葛根颗粒对冠脉结扎大鼠心肌缺血再灌注损伤具有保护作用。
        Objective:To investigate the effect of Shanzha Gegen Granule on myocardial ischemia in rats undergoing coronary artery ligation,and to explore its effect on cardiomyocyte apoptosis in rats.Methods:Sixty male SD rats were divided into 6 groups,10 rats in each group,which were sham operation group,model group,Shanzha Gegen Granule low dose group,Shanzha Gegen Granule medium dose group,Shanzha Gegen Granule high dose group,isosorbide dinitrate group. The rat model of myocardial ischemia-reperfusion injury was established. The left anterior descending coronary artery(LAD)was ligated for 30 min and reperfused for 2 h. Serum troponin was detected by immunoassay in rat abdominal aorta. The expression of myocardial tissue in rats was observed by hematoxylin-eosin staining(HE).The expression of Caspase-3 and AIF were induced by immunization group method detection. Results:Compared with the model group,Shanzha Gegen Granule medium dose group,Shanzha Gegen Granule high dose group and the isosorbide dinitrate group showed decreasing trend. The degree of pathological grade of HE staining was significant,and the expression of Caspase-3 in cardiomyocytes was significantly decreased. Attenuation of AIF expression was not statistically significant. Conclusion:Shanzha Gegen Granule has protective effect on myocardial ischemia-reperfusion injury in rats with coronary artery ligation.
