产后方标准汤剂与配方颗粒剂指纹图谱差异分析

摘要
目的建立产后方标准汤剂高效液相色谱法(HPLC)指纹图谱,比较标准汤剂(SD)与配方颗粒(FG)的差异,为产后方的质量评价提供方法。方法制备10批产后方标准汤剂和10批配方颗粒剂,采用HPLC法建立产后方标准汤剂指纹图谱,通过相似度和多元统计分析比较标准汤剂和配方颗粒剂之间的差异。结果标定25个特征峰,指认了4个色谱峰,其中16号峰为阿魏酸,17号峰为毛蕊异黄酮葡萄糖苷,23号峰为毛蕊异黄酮,25号峰为芒柄花素,多数峰来源于益母草、黄芪、当归和川芎。以10批标准汤剂指纹图谱的共有模式作为对照指纹图谱,10批标准汤剂及10批配方颗粒剂的相似度分别为0.994~0.999,0.775~0.901。与标准汤剂相比配方颗粒剂缺少4、10、21、24号峰,二者的16(阿魏酸)、17(毛蕊异黄酮葡萄糖苷)、19号峰差异较大(OPLS-DA中的VIP值>1)。多元统计分析能明显地将二者区分开来。结论本研究指纹图谱体现了产后方标准汤剂的特点,有效地比较了标准汤剂与配方颗粒剂之间差异,可作为产后方质量评价依据。
        Objective To establish High Performance Liquid Chromatography(HPLC) fingerprint of Postpartum Prescription,to compare the differences between the Standard decoction(SD)and the Formula granule(FG),and provide a method for its quality evaluation. Methods Ten batches of SD and 10 batches of FG were firstly prepared.The HPLC method was used to establish the fingerprint of the SD. The differences between SD and FG were compared by similarity and multivariate statistical analysis. Results Twenty five common chromatographic peaks were labeled and 4 peaks were identified. Among them,the 16 th peak was Ferulic acid,the 17 th peak was Calycosin-7-glucoside,the 23 rd peak was Calycosin,and the 25 th peak was Formononetin. Most of the peaks were from Leonuri herba,Astragali radix,Angelicae sinensis radix and Chuanxiong rhizoma. Using the common mode of the SDs' fingerprints as the control fingerprint, the similarities of SDs and FGs were 0.994 to 0.999 and 0.775 to 0.901,respectively. Compared with the SDs,the FGs lacked peaks 4,10,21 and 24. The peaks 16(Ferulic acid),17(Calycosin-7-glucoside)and 19 were significantly different(VIP > 1 in OPLS-DA). Multivariate statistical analysis can clearly distinguish them. Conclusion The fingerprint of this study reflects the characteristics of SD of Postpartum Prescription,and effectively recognizes the differences between the SD and the FG,which can be used as the basis for quality evaluation of Postpartum Prescription.
