不同参数组合电针对炎性痛模型大鼠镇痛效应及中枢内啡肽的影响

摘要
目的:评价不同参数组合电针对炎性痛模型大鼠的镇痛效应,筛选出电针治疗炎性痛的最佳参数组合。方法:42只雄性SD大鼠,随机分为空白组、模型组、2Hz组、100Hz组、120Hz组、2/100Hz组和2/120Hz组。用完全弗氏佐剂(CFA)建立大鼠炎性痛模型,电针干预大鼠患侧"足三里"和"昆仑"穴,于造模前,造模后24h,治疗第1、3、5、7、9、10天检测大鼠右后足机械缩足阈值(PWTs)。用放射免疫分析法(RIA)检测下丘脑β-内啡肽(β-END)与脑啡肽(ENK)含量、脊髓背角强啡肽(DYN)含量。结果:在电针干预后第7~10天,2/100Hz组和2/120Hz组大鼠PWTs不同程度高于其他电针组(P<0.01);各电针组大鼠下丘脑β-END、ENK含量均不同程度高于空白组和模型组(P<0.01,P<0.05),而2Hz电针组大鼠下丘脑ENK含量、脊髓背角DYN含量高于其他电针组(P<0.01,P<0.05)。结论:不同参数组合电针均能减轻大鼠炎性疼痛,炎性痛早期不同参数组合电针镇痛疗效相当,而后期2/100Hz或2/120Hz频率电针疗效更好。中枢内啡肽分泌是电针镇痛起效机制之一。
        Objective: To evaluate the different parameters combinations electro-acupuncture(EA) on analgesia and central endorphins in inflammatory pain model rats and select the best parameter combination of electro-acupuncture treatment for inflammatory pain. Methods: A total of 42 SD male rats were randomly divided into control group(C group), model group(M group), 2 Hz EA group, 100 Hz EA group, 120 Hz EA group, 2/100 Hz EA group and 2/120 Hz EA group. The inflammatory pain model was established by injecting CFA. EA groups were given the electro-acupuncture on Zusanli(ST36) and Kunlun(BL60) of affected sideat at 24 hours before and after modeling. PWTs were detected in the 1, 3, 5, 7, 9, 10 day after the treatment. The α-END and ENK concentration of hypothalamus, as well as DYN concentration of spinal dorsal horn were measured by RIA. Results:Day7 to day10 after electro-acupuncture treatment, 2/100 Hz and 2/120 Hz EA groups PWTs was higher than other EA groups(P<0.01). The α-END and ENK concentration of hypothalamus in all EA groups were higher than C group and M group(P<0.01,P<0.05). The ENK concentration of hypothalamus and the DYN concentration of spinal dorsal horn in 2 Hz EA group was higher than the other groups(P<0.01, P<0.05). Conclusion: Different parameter combinations electro-acupuncture has analgesic action for inflammatory pain modal rat. At early stage of inflammatory pain, different parameter combinations electro-acupuncture is similar efficacious, while at late stage of inflammatory pain, 2/100 Hz or 2/120 Hz electro-acupuncture is more efficacious. The secretion of central endorphins is one of the effective mechanisms of electro-acupuncture analgesia.
