栽培肾茶遗传多样性的SRAP标记分析

摘要
应用SRAP分子标记技术对云南、广西、福建、海南等地收集的16个栽培肾茶种质进行遗传多样性分析。15对引物经PCR扩增后共检测到124个条带,其中多态性条带80个,多态性条带百分率(PPL)为66.7%;肾茶种质间的观测等位基因数(Na)为1.669 4、有效等位基因数(Ne)为1.434 0,Nei's基因多样性指数(H)为0.252 9,Shannon信息指数(I)为0.374 7,遗传相似度介于0.566 9到0.991 7之间,说明肾茶的遗传多样性水平较低。UPGMA聚类结果将肾茶种质分为2类,与根据花冠颜色对肾茶类型进行划分的结果对应,而遗传特性与地理分布没有明显相关性。实验结果可为肾茶遗传研究、品种选育与资源保护提供科学依据。
        The genetic diversity of 16 cultivated Clerodendranthus spicatus germplasm, collected from Yunnan,Guangxi, Fujian and Hainan province, was analyzed by SRAP molecular marker technology. A total of 124 sites were detected after amplification with 15 pairs of primer combinations, with the number and percentage of polymorphic loci being 80 and 66.67%. Among the germplasm resources of Clerodendranthus spicatus, the number of Na, Ne, H and I were 1.669 4, 1.434 0, 0.252 9 and 0.374 7, respectively, the similarity coefficient ranged from0.566 9 to 0.991 7, which showed a low-level genetic diversity of C. spicatus. C. spicatus germplasm resources were classified into 2 categories based on the UPGMA cluster analysis, which corresponded with the classification by the color of corolla, meanwhile, hereditary characters were not related to the geographical distribution. The results could provide scientific basis for the genetics research, variety breeding and resource conservation of C.spicatus.
