摘要
[目的]分析延边地区朝鲜族和汉族小学生不同代谢指标与磷酸烯醇式丙酮酸羧激酶1(PCK1)基因单核苷酸多态性(SNPs)的相关性.[方法]于2011年8月—2013年8月间在延边地区选择3所小学校,随机抽取1 206名小学生(朝鲜族706名,汉族500名)作为研究对象,测量体质量指数(BMI)、腰围(WC)、收缩压(SBP)和舒张压(DBP);检测空腹12 h血清三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、血糖(FBG)和胰岛素(FIns)值;提取DNA并检测PCK1基因SNPs rs1042531位点的基因型.[结果]朝鲜族小学生PCK1基因SNP rs1042531位点TT,TG及GG型的频率分别为59.5%,36.0%和4.5%,汉族小学生分别为61.6%,33.2%和5.2%,朝鲜族和汉族小学生基因型分布频率间差异无统计学意义(P=0.567);朝鲜族小学生T和G等位基因频率分别为77.5%和22.5%,汉族小学生分别为61.6%和38.4%,朝鲜族和汉族小学生的等位基因频率分布间差异有统计学意义(P<0.01).PCK1基因SNP rs1042531位点不同基因型朝鲜族小学生HDL-C平均水平间差异有统计学意义(P<0.05),汉族小学生BMI,WC,SBP,DBP,FBG平均水平间差异均有统计学意义(P<0.05);朝鲜族小学生PCK1基因SNP rs1042531位点不同等位基因HDL-C平均水平间差异有统计学意义(P<0.01),汉族小学生BMI,WC,SBP,DBP,FBG平均水平间差异有统计学意义(P<0.01).[结论]延边地区朝鲜族和汉族小学生PCK1基因rs1042531位点具有SNPs,等位基因频率分布特征存在民族差异;不同代谢指标与PCK1基因SNPs的相关性有所不同,且存在民族差异.
        OBJECTIVE To analyze the correlation between the different metabolic indexes and single nucleotide polymorphisms(SNPs) of phosphoenolpyruvate carboxykinase 1(PCK1) gene in the pupils of Korean and Han nationality of Yanbian area. METHODS 1 206 elementary school students(706 Korean nationality, 500 Han nationality) were randomly selected from 3 primary schools in Yanbian area from August 2011 to August 2013. The body mass index(BMI), waist circumference(WC), systolic blood pressure(SBP) and diastolic blood pressure(DBP) were measured, blood triglyceride(TG), high density lipoprotein cholesterol(HDL-C), fasting blood glucose(FBG) and insulin(FIns) were measured after 12 hour fasting, DNA was extracted and genotypes of PCK1 gene SNP rs1042531 site were detected. RESULTS The TT, TG and GG genotype frequencies at the PCK1 gene SNP rs1042531 site were 59.5%,36.0% and 4.5% in the Korean nationality pupils, and 61.6%, 33.2% and 5.2% in the Han nationality pupils, respectively, and there was no significant difference in genotype frequency between Korean and Han nationality pupils(P=0.567). The T and G allele frequencies of Korean nationality pupils were 77.5% and 22.5%, and those of Han nationality pupils were 61.6% and 38.4%, respectively, and there was significant difference in the distribution of allele frequencies between the two nationality pupils(P<0.01). The difference of mean levels of HDL-C in the Korean nationality pupils with different genotype of PCK1 gene SNP rs1042531 site was statistically significant(P<0.05), and there were statistically significant differences in BMI, WC, SBP, DBP and FBG of Han nationality pupils(P<0.05). The difference of mean levels of HDL-C in the Korean nationality pupils with different allele frequencies was statistically significant(P<0.01), and there were statistically significant differences in BMI, WC, SBP, DBP and FBG of Han nationality pupils(P<0.01). CONCLUSION The PCK1 gene rs1042531 had SNPs in Korean and Han nationality pupils in Yanbian area, and there were nationality differences in allele frequency distribution. The correlation between different metabolic indexes and SNPs of PCK1 gene was different, and there were nationality differences.
