海河流域溶藻菌Q1的溶藻效果分析及鉴定

摘要
从天津市海河富营养化的水域中分离到1株具有溶藻能力的细菌,将其命名为Q1,对该菌株进行生理生化鉴定及提取16SrRNA分子鉴定,将Q1以10%接种量接入铜绿微囊藻(Microcysis aeruginosa)、水华鱼腥藻(Anabaena flosaquae)和斜生栅藻(Scenedesmus obliquus)的纯培养藻液中,研究Q1对不同微藻的专一性及不同生长时期Q1的菌液对铜绿微囊藻生长的影响、作用方式。结果表明:菌株Q1属于苏云金芽孢杆菌属(Bacillus thuringiensis),该菌株稳定期的滤液溶藻效果最佳,菌株Q1能溶解水华鱼腥藻和铜绿微囊藻,对斜生栅藻也有促进作用。该菌株的无菌滤液经高温处理后,对铜绿微囊藻的溶藻效果没有影响,推断菌株Q1分泌的可耐高温的胞外物质。该菌株与藻体细胞相互抑制,存在竞争共栖的生态关系。
        An algae-lysing bacterium strain named Q1 was isolated from eutrophic waters in the Haihe river of Tianjin.The biophysics,biochemical and the analysis of its 16 SrRNA sequence were conducted.To investigate the specificity of algicidal activity against Microcysis-aeruginosa,Anabaena flosaquae and Scenedesmus obliquus by bacterial Strain Q1,we added Q1 to monoculture system of these three microalage at the rate of 10%.The strain Q1 from different growth periods were used to study its inhibition and mode of action against Microcysis aeruginosa.The numbers of cell and the photosynthetic pigments content in M.aeruginosa.were inve stigated The results showed that the strain Q1 was identified as Bacillus thuringiensis.Q1 on stable growth phase showed the best alga-lysing performance.The strain could effectively dissolve M.aeruginosaand A.flosaquae and promote the growth of S obliquus.The aseptic filtrate of the strain was not affected by M.aeruginosa when it was treated at high temperature.Therefore,it can be concluded that a high temperature extracellular substance secreted by strain Q1 had the characteristics of algae lysis.The interaction between strain and algal cell was mutually inhibited,and there was an ecological relationship of competition and cohabitation,between strain and algal.
