摘要
目的:采用星点设计—响应面法进行试验设计,建立模型优化南烛叶总黄酮的提取工艺,获得最佳提取条件。方法:该实验以芦丁作为成分指标测定南烛叶提取液中总黄酮含量;以提取温度、液料比、乙醇浓度作为自变量,黄酮得率作为响应值,进行星点设计试验,通过design-expert软件建立模型,分析单因素和交互项对南烛叶提取液中总黄酮得率的影响。结果:经design-expert对各因素进行多元二次拟合所得方程为Y=-137.93+2.62A+1.32B+1.73C-4.72×10~(-3)AB-8.25×10~(-3)AC-5.10×10~(-3)BC-0.02A2-8.83×10~(-3)B~2-8.68×10~(-3)C~2,通过模型进行最佳提取工艺的优化,结合实际操作的调整,提取温度46℃、液料比45:1、乙醇浓度65%为最佳提取条件,平行试验3次的总黄酮得率与预测值的偏差为-1.15%。结论:星点设计—响应面方法使用简便,对实际生产过程中提取条件的优化可得到很好的预测结果。
        Objective:To optimize the extraction process of flavonoids from Vaccinium bracteatum Thunb leaves by using central composite design response surface method(CCD-RSM). The optimum extraction conditions were obtained. Methods:This experiment used rutin as a component index for the determination of the total flavonoidsin extract of Vaccinium bracteatum Thunb leaves. Extraction temperature,solid-to-liquid ratio and ethanol concentration were as independent variables and the yield of flavonoids were as the response value of central composite design. The test model was established by designexpert software,analyzing the influence of single factor and interaction on the extraction yield of flavonoids from Vaccinium bracteatum Thunb leaves. Results:Considering each factor in this experiment,a multiple quadratic equation was formulated by design-expert as follows:Y=-137.93+2.62A+1.32B+1.73C-4.72×10~(-3)AB-8.25×10~(-3)AC-5.10×10~(-3)BC-0.02A~2-8.83×10~(-3)B~2-8.68×10~(-3)C~2. To optimize the best extraction process through the model,combined with the adjustment in actual operation. Extraction temperature 46 ℃,solid-to-liquid ratio 45∶1,ethanol concentration of 65% were proved to be the best extraction conditions.During the 3 times parallel tests,the deviation of the obtainedrate of flavonoids and the predictive value was -1.5%. Conclusion:The central composite design response surface method(CCD-RSM)is simple and convenient,and it can be used to optimize the extraction conditions in actual production process.
