骨肉瘤中PTEN蛋白和Skp2表达的相关性研究

摘要
目的通过研究骨肉瘤组织中PTEN蛋白,Skp2的表达水平及两者之间的相互关系,探讨它们在骨肉瘤发生发展的意义。
     方法用免疫组织化学S-P法检测PTEN蛋白、Skp2、p27在30例骨肉瘤和10例骨软骨瘤中的的表达,采用统计学软件SPSS13.0进行统计学分析。
     结果1)骨肉瘤中PTEN蛋白的阳性表达率53.3%,显著低于骨软骨瘤中PTEN蛋白的表达率90.0%(p<0.05)。2)骨肉瘤中Skp2蛋白的表达率76.7%显著高于骨软骨瘤中Skp2蛋白的表达率10.0%(p<0.05)。3)在骨肉瘤组织中,p27的表达率为56.7%。4)在骨肉瘤外科分期间PTEN蛋白,Skp2蛋白阳性表达无统计学显著性差异(p>0.05)。5)在骨肉瘤外科分期中,Ⅲ期骨肉瘤p27蛋白的阳性表达率与Ⅱ期(ⅡA期、ⅡB期)组相比明显下降,有统计学显著性差异(p<0.05),而ⅡA期与ⅡB期骨肉瘤的p27蛋白的表达率没有显著性差异(p>0.05)。6)在骨肉瘤中,Skp2蛋白与PTEN蛋白表达呈负相关(r=-0.47,p<0.01),Skp2蛋白与p27蛋白的表达亦呈负相关(r=-0.82,p<0.01),而PTEN蛋白与p27蛋白的表达呈正相关(r=0.69,p<0.01)。
     结论首次发现在骨肉瘤中PTEN蛋白与Skp2的表达呈负相关。提示在骨肉瘤发生与发展中可能存在一条PTEN-PI3K-Skp2-p27调控途径,该途径通过介导p27的泛素化降解,促使细胞周期调控发生紊乱,导致骨肉瘤的发生发展。
Obiective To investigate the expression and clinical significance of Skp2 and PTEN in osteosarcoma and the correlation between them.
     Method The expression of PTEN,Skp2 and p27 were detected came from 30 cases of osteosarcoma and 10 cases of osteochondroma by SP immunohistochemistry, then we analyzed the result by SPSS 13.0.
     Results 1) PTEN protein expression rate in osteosarcoma was significantly lower than that of osteochondroma in the rate of PTEN protein expression (p < 0.05). 2) The Skp2 protein expression in osteosarcoma was significantly higher than osteochondroma Skp2 protein expression (p <0.05). 3) p27 expression rate was 56.7% in osteosarcoma. 4) In the surgical staging of osteosarcoma, the PTEN protein, Skp2 protein expression was no significant difference(p>0.05)5) In osteosarcoma, the expression of p27 in stage III was significantly different(p<0.05) comparing to the expression in stage IIA、IIB, and stage IIA and IIB with osteosarcoma of the p27 protein expression was no significant difference (p> 0.05). 6) The Skp2 protein and PTEN protein expression was negatively correlated (r = -0.470, p <0.01), Skp2 protein and p27 protein expression was also negatively correlated (r =-0.820, p < 0.01), and PTEN protein and p27 protein expression was positively correlated (r = 0.690, p <0.01)in osteosarcoma.
     Conclusions The negative correlation of PTEN and Skp2 protein expression was first discovered in osteosarcoma.It may be exist of a PTEN-PI3K-Skp2-p27 control channels suggest in the occurrence and progress of osteosarcoma. The degradation of p27 in the ubiquitinpm teasome system was promoted.loss of cell cycle regulation, thus contributing to the occurrence and development of osteosarcoma.
