重症肌无力患者骨髓单个核细胞中细胞周期蛋白D表达的初步观察

摘要
目的:观察重症肌无力(Myasthenia gravis,MG)患者骨髓单个核细胞中细胞周期蛋白D(Cyclin D)不同亚型的表达。
     方法:重症肌无力组10例及健康对照组5例,采集骨髓标本,用淋巴细胞分离液(Ficoll-paque)分离骨髓单个核细胞,按Trizol-氯仿-异丙醇一步法提取细胞总RNA,用反转录PCR(Reverse polymerase chain reaction,RT-PCR)方法检测Cyclin D不同亚型(D1、D2、D3)的表达。
     结果:在重症肌无力组和健康对照组中,Cyclin D1都没有表达;Cyclin D2、Cyclin D3的表达,在重症肌无力组和健康对照组之间无明显差别(p﹥0.05)。
     结论:本研究未发现重症肌无力患者骨髓单个核细胞Cyclin D的表达异常。
Objective: To observe the expression of Cyclin D in the bone marrow mononuclear cells of Myasthenia Gravis(MG) patients.
     Methods: The bone marrow samples were collected from ten MG patients and five normal controls.The bone marrow mononuclear cells were isolated with Ficoll-paque. The total RNA was extracted according to the method of Trizol-chloroform-isopropyl alcohol.The mRNA expres- sion levels of Cyclin D isoforms were detected by semi-quantitative RT-PCR.
     Results: The Cyclin D1 mRNA could not be detected in the bone marrow mononuclear cells of MG patients and normal persons, but the Cyclin D2 and Cyclin D3 could be detected.And the expression levels of Cyclin D2 and Cyclin D3 in MG patients and normal controls show no significance. (p﹥0.05)
     Conclusions: The expression levels of Cyclin D in MG patients are as the same as those in normal controls shows that there is no significant cell phase transition disorders at the G1 phase in our study.
