黄连解毒汤吸收入血有效成分的筛选及生物学作用研究

摘要
研究目的:以复方吸收入血理论为指导,以具有广泛药理作用的中药复方黄连解毒汤大鼠灌服后,从血清中分离出来的黄芩苷、汉黄芩苷、黄芩素、汉黄芩素和栀子苷5个单体成分及由它们组成的混合成分作为主要研究对象,在同一条件下进行了其抑菌、抗炎、对机体免疫影响的活性作用初步研究,确认和筛选出该中药复方活性成分同时,初步研究了该复方中各吸收入血单体有效成分及混合成分的生物学活性,为完善黄连解毒汤临床方面的应用提供依据。
     内容及方法:按复方吸收入血研究方法的要求制备和分析了供试样品。用抑菌试验研究不同供试样品对金黄色葡萄球菌等8个菌的抑菌作用;在对机体免疫影响方面用从形态学上研究了各供试样品对小鼠免疫器官(胸腺和脾脏)发育的影响。细胞水平上研究了吸收入血成分对腹腔巨噬细胞的吞噬功能和对T淋巴细胞转化的影响。在抗炎方面以人组织瘤细胞THP-1产生细胞因子为模型研究了吸收入血成分对THP-1表达IL-1β、TNF-α的影响。
     结果:在抑菌方面,黄连解毒汤吸收入血混合成分及5种单体成分均有抑菌作用,但存在差别,各单体成分的协同作用使黄连解毒汤入血混合成分的抑菌能力强于各单体成分。黄连解毒汤入血成分对小鼠的胸腺、脾脏脏器指数的影响与空白对照相比无显著差异,但在形态学上黄连解毒汤吸收入血成分(黄芩苷除外)对小鼠免疫器官的发育有抑制作用。黄连解毒汤吸收入血成分均增强了腹腔巨噬细胞的吞噬功能。黄连解毒汤入血的黄芩苷促进了T淋巴细胞的转化,而其它各成分抑制了T淋巴细胞的转化。在抗炎方面,各样品均能抑制THP-1细胞表达IL-1β、TNF-α,以汉黄芩素的作用最为显著。
     结论:黄连解毒汤入血混合成分和各吸收入血单体成分均有不同程度的抑菌和抗炎作用。前者的抑菌、抗炎作用更强。黄连解毒汤入血混合成分及5种吸收入血的单体成分均具有免疫调节作用。黄连解毒汤吸收入血成分中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、栀子苷是有效成分。
Objective: In research on the pharmacological activities of Huang Lion Jie Du Tang (HLJDT)--a traditional Chinese compound preparation-the dose were feeded to rats and abstract absorbed components of HLJDT from the serum of the rats under the direction of absorbed theory. By doing that we prepared a great deal of samples containing baicalein,baicalen ,wogonoside , wogonin and geniposide and the total of them. During the bioactivity experiment of inhibit bacteria ,anti-inflammatory ,regulating the immune under the same conditions ,we studied the bioactivity effects of the prepared dose to confirm its pharmacological activities ,select and analysis the activities of the components ,discuss the application of the HLJDT in clinical course and provide the material for the further pharmacological study .
    Contents and methods 'we prepare and analysis the samples according to the absorbed theory .To test the inhibit effects about 8 strains bacteria ,we added the samples to the plates cultured with bacteria,to study the immunoregulating effects of the samples ,we feeded samples to mouse and observed the weight and the structures of its Thymus Gland and Spleen ,the macrophage cell and T lymphocyte's function affected by baicalein,baicalen ,wogonoside , wogonin and geniposide and the total of them. In studying the anti-inflammatory mechanism, we stimulate
    THP-1 -a model for test the ability of produce cytokines, with LPS 10 u g /ml+PMA 200
    nmol/ml, the production of the cytokine reach a peak . The effects of the samples regulating TNFa and IL-1B produced by THP-1 were studied by enzyme-linked immunosorbent assay (ELISA).
    Results : In inhibiting bacteria experiments ,all the samples suppressed the multiplication of the bacteria in different degrees and the sample contain all components makes a significant suppressed effect . The results of observing Thymus Gland and Spleen of mouse showed the samples did not affect the developing of Thymus Gland and Spleen weight, but suppressing the multiplication of the lymphocytes in it except baicalein .the samples increasing the swallow ability of the Macrophage cell and all the samples except baicalein suppressed the T lymphocyte's developing course .All samples inhibited the production of TNFa and IL-1B by THP-1 was stimulated LPS and phorbo-lmyristate acetate (PMA),and it is that significantly inhibit the production of TNFa and IL-1B.
    
    
    
    Conclusion : The effective components absorbed into blood in HLJDT can inhibit bacteria ,perform anti-inflammatory ,regulat immune in different degrees, geniposide, baicalein,baicalen ,wogonoside and wogonin are bioactivity components in HLJDT.
