消渴丸对人血管内皮细胞增殖及一氧化氮产生的影响

摘要
目的:
     1.研究消渴丸不同组分对人血管内皮细胞增殖过程的影响;
     2.研究消渴丸不同组分对人血管内皮细胞NO产生的影响;
     3.研究消渴丸不同组分对人血管内皮细胞eNOS mRNA表达水平的影响。
     通过上述研究,探讨消渴丸不同组分对人血管内皮细胞的作用,以期明确消渴丸中药组分和西药组分对人血管内皮细胞的作用,从而进一步完善消渴丸应用的适应证。
     方法:
     1.应用不同含药血清作用于人脐静脉内皮细胞,使用XTT比色法测定细胞增殖情况;
     2.应用不同含药血清作用于人脐静脉内皮细胞,吸取培养液使用Griess法检测总NO产生情况;
     3.应用不同含药血清作用于人脐静脉内皮细胞,提取RNA,采用荧光实时定量RT-PCR法检测eNOS mRNA表达水平。
     结果:
     1.XTT比色法显示消渴丸和消渴丸中药成分对人脐静脉内皮细胞的增殖能力无显著影响;
     2.总NO检测法提示消渴丸、消渴丸中药组分和格列本脲均对人脐静脉细胞NO产生有促进作用;
     3.荧光实时定量RT-PCR法显示消渴丸(?)组分对人脐静脉内皮细胞eNOS mRNA的表达起促进作用,但没达到显著差异。
     结论:
     本研究首次发现消渴丸和消渴丸中药组分对人脐静脉内皮细胞的增殖无显著影响;消渴丸、消渴丸中药组分和格列本脲均对人脐静脉内皮细胞NO产生有促进作用;消渴丸中药成分对人脐静脉内皮细胞eNOS mRNA的表达起促进作用,但没达到显著差异。
Objective:
     1. To investigate the effects of different parts of the traditional Chinese medicine XiaoKeWan on the proliferation of HUVEC;
     2. To investigate the effects of different parts of the traditional Chinese medicine XiaoKeWan on the production of NO of HUVEC;
     3. To investigate the effects of different parts of the traditional Chinese medicine XiaoKeWan on eNOS mRNA expression of HUVEC.
     This study was aimed at characterizing the response to XiaoKeWan of HUVEC, in terms of proliferation and production of NO, to clarify the function of XiaoKeWan on the endothelial cells and then discuss its therapeutic use from this point.
     Methods:
     1. HUVECs were cultured in the presence of different parts of the traditional Chinese medicine XiaoKeWan. XTT assay was applied to evaluate the cell growth.
     2. HUVECs were cultured in the presence of different parts of the traditional Chinese medicine XiaoKeWan. Then the total NO in the medium was measured with the Griess method.
     3. Realtime PCR was applied to measure the levels of eNOS mRNA expressing in the culture of different parts of the traditional Chinese medicine XiaoKeWan.
     Results:
     1. XiaoKeWan and its Chinese medicine part exhibited no obvious effect on the proliferative activity of HUVEC.
     2. XiaoKeWan, the Chinese medicine.part of XiaoKeWan, and glyburide all increased the production of NO.
     3. The Chinese medicine part of XiaoKeWan increased the expression of eNOS mRNA, but no significant differences.
     Conclusion:
     Our study discovered that XiaoKeWan and its Chinese medicine part exhibited no obvious effect on the proliferative activity of HUVEC; XiaoKeWan, the Chinese medicine part of XiaoKeWan, and glyburide all increased the production of NO; the Chinese medicine part of XiaoKeWan increased the expression of eNOS mRNA, but no significant differences.
