中药穴位敷贴对失眠大鼠下丘脑单胺类递质和IL-1β的影响

摘要
目的:1.观察中药穴位敷贴是否具有镇静催眠作用;2.探讨穴位敷贴治疗失眠的机理;3.比较透皮控释纳米化中药贴片(以下简称CRCP)与传统的中药药贴(以下简称TCMP)穴位敷贴的镇静催眠作用。
     方法:1.测定戊巴比妥钠致眠大鼠睡眠时间实验方法,观察对照组、TCMP组、CRCP组大鼠的睡眠时间的变化;2.将SD大鼠30只随机分为5组:对照组、模型组、安定组、TCMP组、CRCP组,每组6只,以腹腔注射对氯苯丙氨酸(PCPA)建立失眠大鼠模型,中药穴位敷贴于失眠大鼠神门、三阴交穴,以荧光分光光度分析法(FS)和酶联免疫吸附试验(ELISA)为检测方法,观察大鼠下丘脑5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)、多巴胺(DA)、去甲肾上腺素(NA)以及细胞因子白细胞介素1β(IL-1β)的含量变化。3.用Morris水迷宫检测中药穴位敷贴对失眠大鼠的学习记忆能力的影响。
     结果:1.CRCP组和TCMP组与对照组比较戊巴比妥钠致眠大鼠睡眠时间明显延长(P<0.01);2.PCPA腹腔注射后,大鼠正常的睡眠-觉醒周期消失;治疗后失眠大鼠的睡眠-觉醒周期基本恢复。3.PCPA腹腔注射后,失眠大鼠下丘脑5-HT、5-HIAA含量明显下降,与对照组比较有明显差异(P<0.05),而NA、DA、IL-1β含量变化与对照组比较均无明显差异(P>0.05);4.安定治疗后,PCPA所致失眠大鼠下丘脑5-HT、5-HIAA及NA、DA、IL-1β均无明显变化(P>0.05)。5.CRCP组和TCMP组大鼠下丘脑5-HT、5-HIAA、DA、IL-1β含量均变化明显,与模型组比较有非常显著性差异(P<0.01),而TCMP组NA含量与模型组比较无明显变化(P>0.05),CRCP组NA含量与模型组比较有非常显著性差异(P<0.01);5.各组大鼠学习记忆情况随时间变化,CRCP组和TCMP组相同时点大鼠记忆能力相对提高并趋向于对照组方向,与模型组比较有明显差异(P<0.05)。
     结论:1.中药穴位敷贴具有镇静催眠作用,CRCP镇静催眠作用优于TCMP;2.中药穴位敷贴治疗失眠的机制可能是通过调节下丘脑5-HT、5-HIAA、DA、NA、IL-1β等而改善睡眠;3.中药穴位敷贴能够改善失眠大鼠的学习记忆能力。
Objective:
     1. To observe if the Chinese Medicine Patches with acupoint application method has mitigate and hypnotic effect.
     2. To investigate the mechanism of acupiont application method for the treatment of insomnia.
     3. To probe into the controlled-released-nanometer-Chinese medicine patches(CRCP) of acupoint application method better or worse than the traditional Chinese medicine patches(TCMP) of acupoint application method for the treatment of insomnia.
     Methods:
     1. The sleep time of Pentobarbital sodium rats determine method was used to observe the change of sleep time of rats in control group,TCMP group,CRCP group.
     2. Thirty SD rats were randomly divided into five groups and every group had six rats: control group, model group, diazepam group, TCMP group, CRCP group. PCPA abdominal injection rats were used as the models of insomnia in the experiments. Chinese medicine patches of acupoint application on Shenmen(HT7) and Sanyinjiao(SP6).Monoamine Neurotransmitter such as 5-Hydrroxytryptamine(5-HT), 5-Hydroxyindole Acetic Acid(5-HIAA), Dopamine(DA), L-Noradrenaline(NA) and Cytokines such as Interleukin 1β(IL-1β) in the hypothalamus of insomnia rats were observed by Fluorescence spectrophotometry(FS) and Enzyme-Linked Immunosorbent Assay(ELISA).
     3. Morris Water Maze was used to test the influence of Chinese Medicine Patches with acupoint application method on learning and memory ability of insomniac rats.
     Results:
     1. Compared with control group, the sleep time of Pentobarbital sodium rats was obviously prolonged in CRCP group and TCMP group(P<0.01).
     2. After PCPA abdominal injection, the normal sleep-wakefulness cycle of rats was disappeared, but after treatment, the cycle recovered.
     3. After PCPA abdominal injection, the content of 5-HT and 5-HIAA in the brain of rats obviously decreased with comparison to control group(P<0.05), but the content of NA, DA,IL-1βdid not change much compared with control group(P>0.05).
     4. After Diazepam administration, the content of 5-HT, 5-HIAA, NA, DA, IL-1βin the brain of insomniac rats caused by PCPA had no obvious change(P>0.05).
     5. After Chinese medicine patches groups treatment, the content of 5-HT, 5-HIAA, DA and IL-1βhad obvious change(P<0.01); while that of NA of TCMP group had no obvious change(P>0.05) and that of NA of CRCP group had obviously change(P<0.01).
     6. The learning and memory ability of rats in every group was changed as time goes by, the memory ability of TCMP and CRCP groups was greatly improved in the same time with comparison to the model group(P<0.05).
     Conclusion:
     1. Chinese Medicine Patches with acupoint application method has mitigate and hypnotic effect. The mitigate and hypnotic effect of CRCP is better than that of TCMP.
     2. The mechanism of Both of Chinese Medicine Patches with acupoint application method for insomnia treatment was due to its good regulation on Monoamine Neurotransmitter such as 5-HT,5-HIAA,DA,NA and Cytokines such as IL-1βin the hypothalamus of insomniac rats.
     3. Chinese Medicine Patches with acupoint application method can recover the learning and memory ability of insomniac rats.
