电针百会、印堂穴对颅脑损伤大鼠脑内Slit2 mRNA的影响

摘要
本论文由文献综述和实验研究两部分组成。
     第一部分文献综述
     文献综述分为两篇。第一篇介绍了针灸对颅脑损伤作用的临床和实验研究概况。第二篇论述了轴突导向因子Slit家族的研究概况,主要从Slit家族的发现,Slit的结构、分布及表达,Slit的作用几方面进行了论述。同时从Slit2的主要功能和应用两方面论述了Slit2的研究现状。
     第二部分动物实验
     目的:通过研究电针对颅脑损伤大鼠脑内Slit2表达的影响,探讨督脉电针对颅脑损伤的作用机制。
     方法:用改良的Feeney's自由落体打击法建立大鼠中度颅脑损伤模型,将96只大鼠随机分为空白对照组、模型对照组、电针治疗组三个组别,打击后利用神经功能损伤程度评分(Neurological Severity Score, NSS)简易评估大鼠颅脑损伤后的神经功能,且各组大鼠分别于损伤后6小时、24小时、48小时、6天取大脑伤侧顶叶皮层区进行总RNA的提取,经RT-PCR反应、电泳成像与数据分析等过程,测定损伤后伤侧脑组织内slit2 mRNA的表达。
     结果:(1)模型对照组各时间段的神经功能损伤程度评分比空白对照组都高,差异极显著,其P值都小于0.01;电针治疗组各时间段的神经功能损伤程度评分比模型对照组都低,6h组差异极显著(P<0.01),24h、48h、6d组差异显著(P<0.05)。(2)与空白组比较,各时间段模型对照组和电针治疗组大鼠损伤脑组织中Slit2mRNA基因表达升高显著,有极显著性差异(P<0.01);与模型对照组比较,电针治疗组的Slit2mRNA基因升高较明显,有显著性差异(P<0.05)。且在四个时间段中,Slit2 mRNA的表达呈一过性升高而后降低的趋势,且48h达到峰值。结论:电针(督脉)对颅脑损伤的治疗作用可能是通过升高轴突导向因子Slit2的含量,从而参与损伤后神经元的再生和组织修复,改善神经功能,达到电针(督脉)对颅脑损伤组织的保护作用,其机制可能与Slit2介导轴突生长和神经元迁移有关。
literature review and experimental study are two components of the paper.
     First Part:Literature review
     The first review is involved in the clinical and experimental study of the acupuncture and moxibustion on the brain injury; The second review is involved in progresses in research work of the axon guidance factor-Slit family from several aspects such as the discovery, structure, distribution and function; meanwhile, the main function and apply of Slit2 is discussed.
     Second part:Experimental study
     Objective:We studied the electroacupuncture influence to the express of Slit2 in the injuried brain, in order to discuss its mechanism action of acupuncturing governor meridian to the injuried brain.
     Methods:A modified Feeney's free-fall model was used to build the moderate injuried model of the rat We divided 96 rats into three groups randomly:Blank Group, Model Group, and EA Group. After the hit we use the Neural injury severity score (NSS) to evaluate the neurological function on rats. And at 6h,24h,48h, 6d after injury we took the brain tissues to measure the express of Slit2. Also, total RNA was extracted through RT-PCR..
     Results:(1)The NSS in Model Group was significant higher at 6h,24h,48h,6d when it was compared with the Blank Group,the statistical datas of P are all less than 0.01; While the NSS in EA Group was significant lower at 6h(P<0.01),24h(P<0.05),48h(P<0.05),6d(P<0.05) when it was compared with the Model Group.(2) Compared with the Blank Group, The Slit2 mRNA content of injury local tissues were increased significantly in EA Group and Model Group at the corresponding times after the brain injury, and the statistical datas have significant difference (P<0.01). Compared with the Model Group, the Slit2 mRNA content of injury local tissues were increased obviously in EA Group, and the statistical datas have obviously difference (P<0.05). Among each time period, the Slit2 RNA content rose at first, then decreased, and it reached the peak in the 48h group.
     Conclusion:EA has therapeutic effect on the brain injury. It maybe through increasing the Slit2 mRNA content to involve in the progress of neuronal regeneration and tissue repairment, Also EA has protective effect on the brain injury maybe through the Slit's functions of mediate axon outgrowth and neuronal migration, from this neuromal function. improves.
