抗郁散对CUMS大鼠血清T_3、T_4、TSH的影响

摘要
目的:通过观察抗郁散对慢性轻度不可预见应激(CUMS)抑郁症模型大鼠行为学及血清T3、T4、TSH的影响,研究抗郁散对抑郁症动物模型HPT轴的影响,从而探讨抗郁散治疗抑郁症的药效学机制。
     方法:采用多种长期慢性不可预见的轻度应激因子复制大鼠抑郁模型,设正常组、模型组、氟西汀组、抗郁散大、中、小剂量组,连续治疗21天,定期测定各组大鼠体重、糖水消耗量及行为学改变,并用放免法测定大鼠血清T3、T4、TSH含量的变化。
     结果:与正常组比较,模型组大鼠体重下降、垂直和水平运动次数减少、糖水消耗量减少,有显著性差异(P<0.05),模型组大鼠血清T4的含量显著升高、TSH的含量升高,有显著性差异(P<0.05);T3的含量降低,无显著性差异(P>0.05)。与模型组相比,抗郁散大、小剂量组和氟西汀组,能使大鼠的行为学明显改善,恢复接近正常组水平,有显著性差异(P<0.05),血清T4值显著降低,TSH值降低,有显著性差异(P<0.05),而T3的值降低,无显著性差异(P>0.05);与模型组相比,抗郁散中剂量组能明显改善大鼠的行为特征,使大鼠血中清T4、TSH的含量显著降低、T3的含量明显升高,差异有显著性(P<0.05)。
     结论:抗郁散可以改善大鼠行为学特征,使血清T3值含量升高、T4、TSH值含量降低,恢复接近正常组水平,其机制可能与抗郁散调节HPT轴功能紊乱有关,为抗郁散在临床上用于抑郁症的治疗提供理论基础。
Objective: Antidepressants scattered through the observation of unpredictable chronic mild stress rat model of depression behavior and serum T3, T4, TSH effects of antidepressants casual study of animal model of depression, the effects of HPTA in order to investigate the bulk Pharmacodynamics mechanism of antidepressants on depression.
     Methods: Using a variety of chronic unpredictable mild stress model of depression in rats, normal group, model group, Fluoxetine group, antidepressants, scattered large, medium and small dose group, 21 days for treatment, periodic determination of each group rat body weight, sugar consumption and behavior change, and RIA determination of serum T3, T4, TSH concentration change.
     Results: Compared with the normal group, model group rats lost weight, vertical and horizontal reduction in the number of sports, to reduce consumption of dessert, there is significant difference (P<0.05), model group, serum T4 levels were significantly increased, TSH levels rise, there is significant difference (P<0.05); T3 levels decreased, no significant difference(P>0.05).Compared with model group, antidepressants scattered large ,medium and small dose group and the Fluoxetine group, can study the behavior of rats markedly improved, the restoration of nearly normal levels, there is significant difference (P<0.05), serum T4 was significantly lowering, TSH values decrease, there is significant difference (P<0.05), and T3 of the values decrease, no significant difference (P>0.05); Compared with model group, antidepressants Powder dosage group can obviously improve the rat's behavioral characteristics, so that blood clearance of T4, TSH levels significantly decreased, T3 levels increased significantly, there was a significant difference (P <0.05 ).
     Conclusions: Antidepressants can improve the dispersion characteristics of rat behavior, so that the value of serum T3 was increased, T4, TSH values decreased to restore near normal levels, the mechanism of antidepressants may be associated with regulation of HPTA scattered dysfunction related to antidepressants scattered on clinical treatment for depression and provide a theoretical foundation.
