玉屏银屑方对湿热证型银屑病动物模型免疫调节作用的实验研究

摘要
目的:1.初探湿热证型银屑病病证结合动物模型的制造方法并对其进行评价。2.观察预防性应用玉屏银屑方对湿热证型银屑病动物模型的症状、体征及免疫功能等方面的影响,证实本方可能通过调节免疫功能来预防或延缓银屑病的复发。方法:1、将实验豚鼠随机分为三组,正常对照组、银屑病组、湿热证型银屑病造模组,其中湿热证型银屑病造模组采用复合多因素造模法,给予豚鼠肥甘饮食加白酒并置于湿热外环境中以制造湿热证的模型,同时用心得安乳剂制造豚鼠银屑病的模型,从而模拟出湿热证型银屑病的病证结合动物模型;通过对模型豚鼠进行症状体征、血脂、肛温及耳背皮肤病理变化的观察及检测,评价造模动物是否成功。2、将实验豚鼠随机分为四组:正常对照组、银屑病组、湿热证型银屑病造模组和玉屏银屑方组,预防组在造模同时预防性应用玉屏银屑方,其他三组与第一部分相同。造模结束后对模型豚鼠进行症状体征及耳背皮肤病理变化的观察及检测,同时借助流式细胞仪检测外周血T淋巴细胞亚群(CD4+/CD8+比值)的变化,运用ELISA法检测各组实验动物IFN-γ/IL-4比值(即Th1/Th2比值),评价各组动物细胞免疫水平的变化,并通过摘取动物胸腺,比较各组的胸腺指数,观察各组动物免疫器官的变化。
     结果:1、湿热证型银屑病组豚鼠同时出现了典型的湿热证及银屑病的症状体征,血脂及肛温水平均显著高于空白组及单纯银屑病组(P<0.01),耳背皮肤病理Baker评分及炎症细胞浸润数亦高于空白组及单纯银屑病组(P<0.01)。2、经预防性应用玉屏银屑方,造模动物与湿热证型银屑病组豚鼠相比,银屑病症状减缓出现,胸腺指数升高(P<0.05),耳背皮肤病理Baker评分及炎症细胞浸润数明显降低(P<0.05),CD4+/CD8+比值及Th1/Th2比值亦降低(P<0.05)。
     结论:1、湿热证型银屑病动物模型既具备了银屑病的病理特征,又符合中医湿热证的证候表现。2、玉屏银屑方可能通过调节豚鼠的免疫器官和T淋巴细胞的免疫水平,达到预防或减缓银屑病发生的作用。
Objective:1. Preliminary damp-heat syndrome psoriasis Syndrome combination ofanimal models of manufacturing method and evaluated.2. Observe the preventiveapplication the Yuping Yinxie square of damp-heat syndrome animal model of psoriasissymptoms, signs and immune function, Confirmed only by regulating the immune functionin patients to prevent or delay recurrence of psoriasis. Methods:1. Experimental guineapigs were randomly divided into three groups, Normal control group, the psoriasis groupand damp-heat syndrome psoriasis model. Damp-heat syndrome type psoriasis mademodule composite multi-factor modeling method, Give guinea pigs Feigan diet plus liquorand placed in a hot and humid outside environment in order to create the model ofdamp-heat syndrome, While propranolol The emulsion manufacturing guinea psoriasismodel,to simulate the damp-heat syndrome psoriasis disease and syndrome animal model;Signs and symptoms, lipids, rectal temperature by guinea pig model and BTE observationand detection of skin disease management changes,and the Evaluation Whether the animalmodel success.2. The experimental animals were randomly divided into four groups,normal control group, psoriasis, psoriasis made module of damp-heat syndrome andYuping Yinxie square group. Prevention group in made die at the same time theprophylactic use of jade screen silver dust party, the other three groups with theexperimental one are the same. After the building was carried out on the model of guineapigs skin symptoms, signs and behind the ears pathology observation and testing, at thesame time by means of flow cytometry instrument detection of peripheral blood Tlymphocyte subsets (the ratio of CD4+/CD8+) of the change, using the ELISA method todetect each group of experimental animals ratio of the IFN-γ/IL-4(i.e. the ratio of Th1/ Th2), evaluation of each animal cell immunity levels change.And by harvesting animalthymus gland, thymus index is calculated,then observe the changes of each buildinganimal immune organs. Results:1. Damp-heat syndrome type psoriasis guinea pigs at thesame time the typical symptoms and signs of damp-heat syndrome and psoriasis, bloodlipids, anal temperature level is significantly higher than blank group and pure psoriasis(P<0.01), Behind the ears skin pathological grading Baker and inflammatory cellsinfiltrating the number is higher than the blank group and pure psoriasis (P<0.01).2.After the prophylactic use of jade screen silver crumbs party, building animals comparedwith damp heat syndrome psoriatic group of guinea pigs, psoriasis symptoms appear slow,thymus index rise(P<0.05), behind the ears skin pathological score and the number ofinflammatory cells infiltrating Baker is decreased obviously (P<0.05), and the ratio ofTh1/Th2and CD4+/CD8+is also lower(P<0.05). Conclusion:1. Damp heatsyndrome psoriatic animal models with both the pathological characteristic of psoriasis,and conform to the damp heat syndrome of TCM syndromes.2. The Yuping Yinxie only byadjusting the level of guinea pig immune organs and immune T lymphocytes to achieve theprevention or slowing psoriasis role.
