铁及其络合物对银屑病治疗作用实验研究

摘要
【目的】
     本研究利用雌激素周期小鼠阴道上皮细胞有丝分裂和鼠尾鳞片表皮颗粒层形成两种银屑病的动物实验模型,探讨铁及络合铁对银屑病的治疗作用。
     【方法】
     1.铁及络合铁对雌激素周期小鼠模型阴道上皮细胞有丝分裂的影响
     使用昆明种雌性小鼠建立雌激素周期小鼠阴道上皮细胞有丝分裂模型,随机分为阴性对照组、阳性对照组,FeCl_3低、中、高剂量组,Fe-EDTA低、中、高剂量组,朴雪口服液组。光学显微镜下观察阴道上皮细胞有丝分裂情况,计算有丝分裂指数。
     2.铁及络合铁对鼠尾鳞状表皮模型颗粒层形成的影响
     将实验小鼠随机分为9组(同上)。光学显微镜下观察鼠尾鳞片表皮颗粒层形成情况,计数100个鳞片中有颗粒层的鳞片数。
     3.模型小鼠组织微量元素含量的测定
     取各组小鼠的肝、脾、肾、尾组织经微波消解仪消解后用原子吸收分光光度计分析小鼠各组织中的铜、铁、锌、钙、镁、锰含量。
     4.对模型小鼠肝、脾、肾组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)的影响
     用试剂盒测定各组小鼠肝、脾、肾组织匀浆中SOD、GSH-Px的活力和MDA含量。
     5.对模型小鼠血清IL-2和IL-10含量的影响
     用放射免疫法测定各组小鼠血清中IL-2和IL-10含量。
     【结果】
     1.铁及络合铁对雌激素周期小鼠模型阴道上皮细胞有丝分裂的影响
     与阴性对照组相比,各补铁组和阳性对照组均显著降低,各补铁组与阳性对照组比较,差异无统计学意义(P>0.05),提示各补铁组对小鼠阴道上皮细胞有丝分裂有显著的抑制作用,其效果与MTX相似(P<0.01)。除阴性对照组外,其他各组进行两两比较,均无统计学差异(P>0.05)。
     2.铁及络合铁对鼠尾鳞状表皮模型颗粒层形成的影响
     各补铁组和阳性对照组与阴性对照组相比均显著升高(P<0.01),各补铁组与阳性对照组无统计学差异(P>0.05),提示各补铁组对鼠尾上皮鳞片中颗粒层形成有显著的促进作用,效果与MTX相似。除阴性对照组外,其他各组进行两两比较,均无统计学差异(P>0.05)。
     3.模型小鼠组织微量元素含量的测定
     脾组织中各补铁组及阳性对照组的铁含量均高于阴性对照组(P<0.05),阳性对照组与各补铁组比较,差异无统计学意义(P>0.05)。在肝、肾、尾组织中,各组铁含量两两比较均无统计学意义(P>0.05)。
     肝、脾、肾、尾组织中,各组Cu、Fe、Mg、Mn、Ca5种元素含量分别两两比较均无统计学差异(P>0.05)。
     4.对模型小鼠肝、脾、肾中SOD、GSH-Px和MDA的影响
     肝组织中,各补铁组SOD活力及GSH-Px活力降低,MDA含量升高,但差异无统计学意义(P>0.05)。脾组织中,各补铁组SOD活力,MDA含量升高,但差异无统计学意义(P>0.05)。肾组织中,各组之间两两比较,SOD活力、MDA含量及GSH-Px活力均无统计学差异(P>0.05)。
     5.对模型小鼠血清IL-2和IL-10含量的影响
     阳性对照组和各补铁组血清中的IL-2含量高于阴性对照组,但差异无统计学意义(P>0.05),各补铁组与阳性对照组比较无统计学意义(P>0.05)。补铁组间两两比较均无统计学意义(P>0.05)。
     阴性对照组、各补铁组血清中的IL-10含量高于阳性对照组,但是无统计义意义(P>0.05)。各补铁组间两两比较均无统计学意义(P>0.05)。
     【结
     1.补铁后雌激素周期模型小鼠阴道上皮细胞有丝分裂受到显著抑制,鼠尾鳞片颗粒层形成显著促进。
     2.补铁后只是脾铁含量显著升高,对必需元素铜、铁、锌、镁、锰和钙无明显促排作用。
     3.补铁可能影响抗氧化体系,提高机体应激水平。
     4.补铁对血清IL-2水平,IL-10水平无明显影响。
Objective
     The effect of iron and iron complex on inhibiting psoriasis development in mouse vaginal epithelium model and mouse tail scale epidermis model were explored.
     Methods
     1.Effects of iron and iron complex on the vaginal epithelium mitosis of estrone periodical mice model.
     Female kunming mice were used to establish the epidermis hyperplasia of estrone periodical mice model.They were randomly divided into nine groups: negative control,positive control,low,middle,high dosage of FeCl_3,low,middle, high dosage of Fe-EDTA and iron lactate.The mice were sacrificed by cervical dislocation after the last administration,and the vaginal mitosis index was observed under optical microscope.
     2.Effects on mouse mouse-tail scale epidermis models
     Mice used as the epidermis parakeratosis model were randomly divided into nine groups indicated as above.The formation of mouse-tail scale epidermis was observed and the numbers of scale with granular layer per 100 scales were examined by optical microscope.
     3.Analysis of essential elements.
     To investigate concentrations of copper,iron,zinc,calcium,magnesium, manganum and selenium,liver,spleen,kidney and tail of mice in all groups were taken and digested to analyze the elements concentration by atomic absorption spectrophotometry.
     4.Effects on recovering the anti-oxidizing system
     The activities of SOD,GSH-PX and the content of MDA in the homogenate of liver,kidney and spleen of the mice were determined in each group.
     5.Effects on the serum level of IL-2 and IL-10
     Radioimmunoassay was applied to measure the serum level of IL-2 and IL-10 in each group.
     Results
     1.Effects of zinc and zinc complex on mouse vaginal epithelium model
     Every iron spiked groups as well as methotrexate group could obviously inhibit the mitosis of vaginal epithelial cells(P<0.01).No statistical significance was found between each iron spiked group and the positive control group(P>0.05).
     2.Effects on mouse tail scale epidermis models
     There was statistical significance between each iron spiked group as well as the positive control and the negative control group(P>0.05).
     3.Effects on removing other essential elements
     Iron contents of each iron spiked group and methotrexate group in spleen were higher than that of the negative control group(P<0.05).No statistical significance was found between the iron spiked group and methotrexate group(P>0.05).In comparison with iron contents,no statistical significance existed among the groups in the liver, kidney and tail(P>0.05).
     In comparison with copper,zinc,magnesium,manganum and calcium contents, no statistical significance existed among the groups in the liver,spleen,kidney and tail(P>0.05).
     4.Effects on recovering the anti-oxidizing system
     In comparison with the corresponding data of the negative control group,the activities of SOD decreased while the contents of MDA increased.However,no statistical significance existed in the livers,kidneys and spleen.
     5.Effects on the serum level of IL-2 and IL-10
     The serum level of IL-2 in each iron spiked group as well as methotrexate group was higher than that in the negative control group,but no statistical significance existed(P>0.05).
     The serum level of IL-10 increased in each iron spiked group,but there was no statistical significance(P>0.05).
     Conclusion
     1.As a result of iron replenishment,the mitosis of vaginal epithelial cells was obviously·inhibited,and the formation of granular layer in mice tail scale was significantly promoted.
     2.Iron replenishment could obviously enhance iron contents in spleen and had no obvious effect on other essential elements such as copper,zinc,magnesium, manganum,and calcium.
     3.It could also enhance the activity of the antioxidant enzyme.
     4.There were no obvious effects on the serum level of IL-2 and IL-10.
