柴胡疏肝散水提取物在大鼠外翻肠囊实验中的吸收研究

摘要
目的:通过建立大鼠外翻肠囊实验模型,考察柴胡舒肝散水提物中6种指标性成分体外肠吸收特征。方法:采用大鼠外翻肠囊实验模型,通过建立UPLC定量分析方法测定不同时间点十二指肠、空肠、回肠中芍药苷、阿魏酸、芸香柚皮苷、柚皮苷、新橙皮苷、甘草酸的肠道吸收量,计算吸收动力学参数,考察上述成分在不同肠段的吸收特征。结果:各肠段累积吸收量(Q)结果显示,除甘草酸外,其他5种成分在各肠段的累积吸收量均具有明显差异;肠吸收速率(Ka)计算结果显示:芍药苷的吸收速率空肠>十二指肠>回肠;阿魏酸、芸香柚皮苷、新橙皮苷的吸收速率十二指肠>空肠>回肠;柚皮苷吸收速率空肠>回肠>十二指肠;甘草酸的吸收速率在空肠吸收好于十二指肠和回肠,但无显著差异。结论:肠囊对柴胡舒肝散水提物中6种指标性成份吸收具有选择性,各成分在不同肠道中的吸收均为线性吸收,其相关系数的平方(R~2)均达到0.9以上,符合零级吸收速率。
OBJECTIVE The research aimed to evaluate the intestinal absorption of extracted by decoction extraction processes from Chaihu-ShuGan-San via everted gut sacs.To investigate the influence of active compounds of Chaihu-Shu-Gan-San(CSGS)decoction extraction on the intestinal absorption of paeoniflorin,ferulic acid,narirutin,naringin,neohesperidin and glycyrrhizic acid,respectively.METHODS:Taking rat everted gut sac model,intestinal sac contents were collected at different time and absorption concentration of the six components above was determined by UPLC,absorption kinetics parameters were calculated for describing their absorption characters in different intestinal segments(duodenum,jejunum and ileum).RESULTS:Absorption of glycyrrhizic acid in duodenum,jejunum and ileum was no significant difference,while the other five components showed large differences;The absorption rate(Ka)show that:the absorption rate of paeoniflorin was jejunum>duodenum>ileum;the absorption rate of ferulic acid,narirutin and neohesperidin was duodenum>jejunum>ileum;the absorption rate of naringin was jejunum>ileum>duodenum;the absorption rate of glycyrrhizic acid in jejunum was better than the other two intestinal segments,while there was no significant difference.CONCLUSION:The absorption of six components of decoction extraction processes from Chaihu-Shu-Gan-San had choices,every component was linearity in different intestine,and the square of coefficient correlation exceed 0.9,which consistent with zero order rate process.
