冷胁迫下二倍体野生种瑟伯氏棉和三裂稀转录组比较分析(英文)

Cotton(Gossypium)is one of the important fiber crops of the world.Cold stress affects the nor-mal growth of cotton in every period.Although it is a complicated mechanism to response cold stress,RNA-Seq technology provides a new strategy to reveal that.The diploid species Gossypium thurberi and Gossypium trilobum exposed to cold stress(4℃)were used to study the cotton cold tolerance mechanisms by the next-generation RNA-Seq technology.Phenotypic and physiological analysis proved that G.thurberi was better at regulating to adapt cold stress than G.trilobum.We mapped RNA-Seq data to reference genome(G.raimondii),then 23 726 and 23 820 genes had been found in G.thurberi and G.trilobum,severally.What is more,there are 4227 and 4174 differentially expressed genes(DEGs)were found in G.thurberi and G.trilobum,respectively.Furthermore,we found signal transduction,transcription factors,and carbohydrate transport and metabolism play important roles in responding to cold stress by DEGs enrichment analysis.And,in this study,we found that G.thurberi accumulated higher levels ofabscisic acid(ABA)than G.trilobum.ABA-mediated CBF4 signal network have been predicted by our data.ICE2,CBF4,RAP2.7,key genes of ABA biosynthesis,that belong to CBF4 regulated network,were identified and analyzed.And,these genes expression were up-regulated because of cold stress.These data suggest that ABA mediated CBF4 regulated network also play a core role in cotton.Therefore,this study not only provides materias and a basic theory for cotton cold tolerance mechanism,but also a new idea to study abiotic stress deeply.
