培养条件对蒿柳花粉萌发及花粉管生长的影响

摘要
采用花粉离体培养法,研究不同的温度、光照及培养基组分对蒿柳花粉萌发的影响。结果表明:培养温度和光照强度均影响蒿柳花粉萌发率和花粉管长度,20~25℃是蒿柳花粉萌发的最适温度区间,1500lx和2000lx是蒿柳花粉萌发的最佳光照强度。正交试验中,一定浓度的蔗糖、H_3BO_3及pH值均可显著影响蒿柳花粉萌发,3个因子最佳组合为蔗糖100g·L~(-1)+H_3BO_3 200mg·L~(-1)+pH5.0,在光照强度为1000lx,温度为25℃时,萌发率高于48.8%。利用试验筛选出的蒿柳花粉最佳离体培养组合培养花粉,并与花粉在柱头上活体萌发相比较,二者花粉管长度无显著差异。
The effects of temperature,light intensity and culture medium composition on Salix viminalis pollen germination and pollen tube growth were investigated with in vitro culture.The results were showed that Salix viminalis pollen germination and tube growth could be affected by cultivation temperature and light intensity.Pollen germination of the optimum temperature range was 15-25 t,and the highest germination rate was under the light intensity of 1500 lx and 2000 lx.Sucrose,H_3BO_3and pH value could promote pollen germination within a certain concentration range remarkably in orthogonal design.The optimal culture medium was sucrose 100 g · L~(-1) + H_3BO_3 200 mg · L~(-1) + pH5.0,and in the medium the rate of pollen germination was higher than 48.8%under the conditions of 1000 lx and 25℃.Pollen germination was conducted on the best combination with in vitro was no significant differences compared with pollen germination on stigma.
