基于卟啉的共价有机框架结构的制备及其在光动力疗法中的应用

摘要
共价有机框架材料是由多种有机单元通过共价键链接而构筑的有机材料,由于构筑基团的多样性、构建反应的多样性及合成方法可控性,共价有机框架材料具有众多优点。卟啉类化合物是一种在人体内具有重要生理功能的光敏分子,并且具有对称的化学结构,被广泛应用于构建多种共轭框架结构并在光动力疗法中具有良好的应用价值。然而这类光敏分子水溶性较差,很难直接注射,并且在肿瘤部分富集效果不佳。我们选取化疗药物顺铂前药作为交联剂,制备了基于卟啉的具有刺激响应性的水溶性共价有机框架材料。在肿瘤细胞内较高还原性的条件下,纳米材料可发生裂解,并且在相应波长的激光照射下,纳米材料可产生活性氧,进一步提高纳米材料对肿瘤的治疗效果。
Covalent-organic frameworks(COFs) are a new class of organic materials which are prepared by conjugation of organic unites, exhibiting great potential in biomedicine due to the diversities of construction groups and reactions as well as the controlled synthesis methods. Porphyrin based compounds, a kind of widely used sensitizer, play the important role in physiological system of human body. However, the commonly used porphyrin based compounds are not only poor in water-solubility but also poor in accumulation at tumor tissue. Herein, the cisplatin prodrug is used as the crosslinker to construct the stimuli-responsible and water-soluble porphyrin based COFs which could degrade under the high reduction lever of tumor cells. Moreover, the reactive oxygen generated from COFs under the irradiation at the specific wavelength will further improve the tumor treatment results.
