养殖半滑舌鳎胃肠道4种酶和黏液的组织化学定位细胞

摘要
研究了半滑舌鳎胃肠道酸性磷酸酶(ACP),碱性磷酸酶(ALP),非特异性酯酶(NSE),过氧化物酶(POD)和黏液细胞的分布。ACP主要分布于胃黏膜上皮细胞的细胞质和肠上皮细胞核上区域和固有层中,且在肠道内的活性比胃强(P<0.05);ALP分布于胃的血管壁,肠的肌层,固有层和上皮细胞的纹状缘和核上细胞质,肠道的酶活性最高,且前肠高于后肠(P<0.05);NSE均匀分布于胃肠道粘膜上皮细胞的细胞质,前肠酶活性显著最高(P<0.05);POD分布于固有层的血细胞中,在胃肠道的强度无明显差异(P>0.05)。AB-PAS组织化学结果显示胃肠道中有三种类型的黏液细胞。Ⅰ型黏液细胞少量分布于胃黏膜上皮细胞间,普遍分布于肠上皮细胞间。Ⅱ型和Ⅲ型黏液细胞仅分布于肠绒毛的中上部上皮细胞间。
The histochemical distribution ofacid phosphatase(ACP), alkaline phosphatase(ALP),non-specific esterase(NSE), peroxidase(POD) as well as the mucous cells in the gastrointestinal tract of Cynoglossus semilaevis was evaluated. ACP was found in the cytoplasm of epithelial cells of the stomach, and in the supranuclear region of enterocytes and the lamina propria of the intestine, the intensity in the intestine was stronger than stomach(P < 0.05). ALP was detected in blood vessels of the stomach, and in muscular layer, lamina propria and striated border and supranuclear cytoplasm of enterocytes in the intestine, the intensity in the anterior intestine was stronger than the posterior side, and the latter was stronger than that in the stomach(P<0.05). NSE was located in the cytoplasm of epithelial cells in whole gastrointestinal tract, the intensity in the anterior intestine was the highest(P<0.05). POD was located in the haemocytes of the lamina propria and the levels were almost similar to the stomach and intestine(P > 0.05).AB-PAShistochemical results revealed three types of mucous cells in the gastrointestinal tract.Type Ⅰ cells were observed among the epithelial cells in the stomach sparselyand enterocytes in the intestie evenly. Type Ⅱ and type Ⅲ cells, were distributed ubiquitously among enterocytes in the middle and top region of intestinal villi.
