草鱼(♀)×赤眼鳟(♂)杂交F_1的可育性研究

摘要
为探讨草鱼(♀)×赤眼鳟(♂)杂交F_1的可育性,本研究以草鱼(♀)×赤眼鳟(♂)杂交F_1及其亲本为材料,采用体外胸腔注射法制备染色体标本,分析和比较了染色体核型,初步解释了杂交F_1同亲本间的遗传关系;同时利用石蜡切片法对其性腺进行了组织学分析,初步评估了杂交F_1的生殖能力和可育潜能。核型结果表明:草鱼、赤眼鳟和杂交F_1的染色体数目均为48条,二倍体,染色体核型公式分别为2n=22m+22sm+4st,NF=92;2n=24m+22sm+2st,NF=94;2n=26m+20sm+2st,NF=94。杂交F_1的性腺结构可分为三种类型:(1)无性腺型(脂肪型):约占40.3%,呈脂肪状,没有或可见少量的类似性腺组织;(2)精巢型:约占45.2%,精巢小叶内有许多精子细胞,但均发育异常,推测为超亲本性成熟或为雄性不育;(3)卵巢型:约占14.5%,性腺中的卵细胞可发育到第Ⅳ时相,能观察到成熟卵子,部分第Ⅳ时相卵母细胞卵黄颗粒稀疏,无法正常排卵,初步判断雌性杂交F_1为部分可育。
The histological section observation and chromosome karyotype were used to explore the fertility of Ctenopharyngodonidellus(♀)×Squaliobarbuscurriculus(♂)F_1.The genetic relationship between the hybrid F_1 and its parents were preliminarily explained by analyzing their karyotypes, moreover, the reproductive capability were evaluated by observing their gonaddevelopment. The karyotype results were as follows: the chromosome number of hybrid F_1,C.idellus and S.curriculus were equal to 48, and all were diploids. The karyotypes of them were2n=22m+22sm+4st, NF=92;2n=24m+22sm+2st, NF=94;2n=26m+20sm+2st, NF=94, respectively.Hybrid F_1 have threegonad types. The first type was free-gonad type(fat tissue type), which accounted for 40.3% of the total. The second type was testis type, which accounted for 45.2%, all spermatids developed abnormally or degenerated. The third type was ovary type, which accounted for 14.5%, among those ovaries,someone can ovulate, but others can't observe matural eggs. So,the phenomenon showed the female hybrid F_1 was fertile partially.
