顺铂+雷帕霉素/3-甲基腺嘌呤混合药物对HeLa细胞自噬的影响

摘要
顺铂是广泛应用于临床癌症治疗的抗癌药物,雷帕霉素和3-甲基腺嘌呤是对细胞自噬具有特异作用的药剂。本文将顺铂分别与自噬诱导剂雷帕霉素和自噬抑制剂3-甲基腺嘌呤混合,探索混合药物对HeLa细胞自噬的影响。分别采用MDC荧光染色、TEM、Western Bloting和MTT等实验技术观察混合药物作用后HeLa细胞自噬囊泡和细胞超微结构的变化、胞内自噬相关蛋白LC3-Ⅰ和LC3-Ⅱ的表达以及药物对细胞的毒性作用。结果显示,随着混合药物浓度的增加,Hela细胞的增值受到抑制,MDC阳性细胞数量增多,胞内自噬体、自噬溶酶体逐渐形成,自噬蛋白LC3-Ⅱ的表达增强且自噬水平上升。这表明顺铂分别与雷帕霉素和3-甲基腺嘌呤按不同比例混合后的药物可显著抑制HeLa细胞生长并诱导细胞发生自噬。
Cis-diamminedichloroplatinum(cisplatin),as a significant chemotherapeutic drug for the treatment of many cancers,has been mixed with rapamycin(RAPA),an autophagy inducer,or 3-methyladenine(3-MA),an autophagy inhibitor,and the mixed medicines have been performed on HeLa cells to explore the specific effects on the autophagy from cell viability assays,transmission electron microscopic observation(TEM),Dansylcadaverine(MDC) staining and Western Bloting analysis.Cisplatin mixed with either rapamycin(RAPA) or 3-methyladenine(3-MA) induces stronger effects on the autophagy than itself does.The mixed medicines inhibite growth in a dose-dependent manner in HeLa cells and subsequently sensitize the cells to apoptosis and death.These experimental investigations can provide useful and fundamental information for cytotoxic chemotherapy efficacy in pharmaceutical and biotechnology fields.
