前列舒丸对慢性非细菌性前列腺炎大鼠血清中TNF-α、IL-1β、IL-2和IL-10的影响

摘要
目的:研究前列舒丸对慢性非细菌性前列腺炎(CNP)大鼠血清中TNF-α、IL-1β、IL-2和IL-10的影响,探讨其治疗CNP的作用机制。方法:成年雄性SD大鼠50只建立CNP模型30d后随机分成5组,模型组、前列舒丸高、中、低剂量组(12、6、3 g/kg)及阳性药组(前列康片,10 g/kg),并以10只正常成年雄性SD大鼠为对照组,给予前列舒丸高、中、低剂量组和阳性药组大鼠灌胃,对照组和模型组20m1/kg蒸馏水灌胃,每天-次,连续30天。30天后处死大鼠取血清和前列腺,光镜观察前列腺组织形态学变化并计算其指数,ELISA法测定血清中TNF-α、IL-1β、IL-2和IL-10含量。结果:对照组前列腺组织正常,未产生炎症,模型组炎症最明显,前列舒丸高、中、低剂量组及阳性药组炎症较轻,炎症有不同程度改善。与对照组相比,模型组前列腺指数降低,血清中TNF-α、IL-1β、IL-2和IL-10水平明显升高,差别有统计学意义(P<0.05);与模型组相比,前列舒丸高、中、低剂量组及阳性药组前列腺指数和TNF-α、IL-1β、IL-2和IL-10水平显著降低,前列舒丸高剂量组降低较阳性药组更明显(P<0.05)。结论:前列舒丸可以减轻CNP的炎症反应,其作用机制可能与降低血清中TNF-α、IL-1β、IL-2和IL-10水平有关。
Objective:To observe the effect of Qianlieshu pill on TNF-α、IL-lβ、IL-2、IL-10 in serum of chronic nonbacterial prostatitis(CNP)rats,and to explore therapeutic:mechanism.Methods:After 30 days the experimental CNP rat model was made.totally 50 rats were randomly divided into 5groups,the model group,the high、 medium、 low dose Qianlieshu pill group(12、6、3 g/kg),the positive medicine group(Qianliekang pill,10 g/kg).Besides,10 normal rats were recruited as a control group.Pill was administered to rats in the positive medicine group and the three dose Qianlieshu pill groups by gastrogavage,Distillded water at 20 ml/kg was administered to rats in the model and control group by gastrogavage,once dally for 30 successive days.After 30 days the rats were killed,serum and Prostates were based.The prostatic index was calculated.Under light microscope,histomorphology changes of rat prostates were observed.The levels of TNF- α、IL- 1β、IL- 2 、IL- 10 in serum in the 6 group rats were measured by ELISA.Results:Comparison with the control group,Prostatic inflammatary in the rats of model grop was induced obviously.Comparison with the model group,Prostatic inflammatory reactions were milder in rats of the positive medicine group and each dose Qianlieshu pill group and their degrees were improved to some extent.Comparison with the control group,the prostate index obviously decreased,The levels of TNF-α,IL- 1 β,IL- 2,IL- 10 in serum significantly increased in the model group(P < 0.05).Comparison with the model group,the prostate index obviously decreased,The levels of TNF-α,IL- lβ,IL-2,IL- 10 in serum significantly decreased in the positive medicine group and each dose Qianlieshu pill group(P < 0.05).Comparison with the positive medicine group,The levels of TNF-α,IL- 1β,IL- 2,IL- 10 in serum significantly decreased more in the high dose Qianlieshu pill group(P <0.05).Conclusion:Qianlieshu pill could treat CNP,which may be associated with lowering levels of TNF-α,IL- 1β,IL- 2,IL- 10 in serum.
