聚(甲基乙烯基醚-alt-马来酸)超分子凝胶的制备和性能

摘要
聚(甲基乙烯基醚-alt-马来酸)(P(MVE-alt-MA))是一种具有良好生物相容性的高分子材料,已经广泛应用于健康保健和医药领域。本工作将应用β-环糊精(β-CD)和偶氮苯(Azo)(或金刚烷Ad)之间的主客体相互作用形成P(MVE-alt-MA)超分子凝胶并研究其外界刺激响应性和应用。首先分别制备主体大分子链(H-P),即β-环糊精接枝的P(MVE-alt-MA)(P(MVE-alt-MA)-g-β-CD),和客体大分子链(G-P),即偶氮苯(Azo)(或金刚烷Ad)接枝的P(MVE-alt-MA)(P(MVE-alt-MA)-g-Azo或P(MVE-alt-MA)-g-Ad),然后将H-P和G-P按照一定的浓度和比例混合即可形成P(MVE-alt-MA)超分子凝胶,P(MVE-alt-MA)-g-β-CD/P(MVE-alt-MA)-g-Azo和P(MVE-alt-MA)-g-β-CD/P(MVE-alt-MA)-g-Ad,并应用二维核磁(2D-NOESY)证实了超分子凝胶的形成。该类P(MVE-alt-MA)超分子凝胶不仅对光刺激响应,而且对pH和温度具有敏感性,并且具有一定的自修复性能。所获得的超分子凝胶也具有良好的细胞相容性,适合作为三维细胞培养支架。
Poly(methyl vinyl ether-alt-maleic acid)(P(MVE-alt-MA)) is a kind of biocompatible material, which is currently widely used in healthcare and biomedical fields. In this work, P(MVE-alt-MA)-based supramolecular hydrogels were prepared through making using of the traditional host-guest interaction of β-CD and azobenzene(or adamantane), and its external-stimulus responsiveness and some application were investigated. Host polymer(H-P), β-cyclodextrin(β-CD)-grafted P(MVE-alt-MA)(P(MVE-alt-MA)-g-β-CD) and guest polymer(G-P) azobenzene(or adamantane)-grafted P(MVE-alt-MA)(P(MVE-alt-MA)-g-azo) or(P(MVE-alt-MA)-g-ad) were first prepared. After simply mixing the aqueous solutions of the host polymer and guest polymer with different concentration and ratios of H-P to G-P, the supramolecular hydrogels P(MVE-alt-MA)-g-β-CD/P(MVE-alt-MA)-g-azo or P(MVE-alt-MA)-g-β-CD /P(MVE-alt-MA)-g-ad were obtained. The formation of the supramolecular hydrogels was also confirmed by 2D-NOESY. This kind of supramolecular hydrogels not only exhibits photo-, pH- and thermo-sensitivity, but also cytocompatibility. P(MVE-alt-MA)-based supramolecular hydrogel is biocompatible, may be expected to have a applications as a three-dimensional(3D) cell culture matrix.
