太平洋大青鲨种群结构初探

根据2011年12月-2013年10月我国远洋延绳钓渔船在太平洋海域(145°-180°W,10°S-15°N)的三个采样区域采集的样本,分析了来自中部太平洋三个采样点的78个个体的序列多样性与种群遗传结构。结果显示,697bp片段中发现了3个变异位点,78个个体出现4个单倍型。序列多样性分析结果显示,78个个体的平均单倍型多样性指数为0.693483,核苷酸多样性指数为0.000995。分子方差分析(AMOVA)显示出现在种群内的遗传变异性为96.06%,群体间的FST分析表明3个采样点大青鲨群体内部并不存在显著的遗传分化,属于一个种群。单倍型遗传距离为0.001-0.003之间,表明该3个区域的大青鲨种群之间存在强烈的遗传交流,种群遗传分化水平较低。而根据2009-2014年太平洋海域(30°S-20°N)中国金枪鱼延绳钓观察员测定的右鳍角长分析,以赤道(0°)划分的南北太平洋群体,两群体之间的右鳍角长存在显著的差异,表明了南北太平洋大青鲨群体在生物学上分属两个相对独立的种群。生物学上的显著差异与Cyt b基因的非显著差异,说明了太平洋大青鲨种群结构并不明朗。在今后的研究中,我们应该结合更多的分子标记技术,微化学分析技术,化石研究以及其他生物学特征等,进行更加深入的研究,探讨太平洋大青鲨的种群结构现状,为大青鲨渔业资源的科学评估和可持续利用提供理论基础。
The blue shark(Prionace glauca) is commercially important fishery species in the world. Assessment of population genetic diversity levels of blue shark is fundamental to the conservation and management of this species. The samples derived from the Chinese vessel were collected from three locations of Pacific waters(145°-180°W,10°S-15°N) from December 2011 to October 2013.We analyzed the sequence variations and population genetic structures for 78 samples collected from 3 locations. In total, 3 variable sites were acquired, and all of them were parsimony sites. 4 haplotypes were identified from 78 individuals..Analysis of mt DNA Cyt b sequences of blue shark from the three localities revealed that the mean haplotypic diversity was0.693483, while the nucleotide diversity was 0.000995. The AMOVA test of Prionace glauca reveales that 96.06% of the genetic variation occurred within populations. FST analysis suggested a non-significant genetic differentiation was found between the three localities and they seem as a whole population.
