蛇麻酮及同系物异源生物合成途径构建

摘要
蛇麻酮及其同系物又称β酸(与α酸合称为苦味酸),是一类异戊烯化的间苯三酚衍生物,主要包含蛇麻酮、类蛇麻酮和聚蛇麻酮,在工业上用于酿造啤酒,同时也具有抗菌、抗氧化、镇静等临床应用价值。蛇麻酮类化合物主要来源于啤酒花,分离纯化成本高,其大规模制备还受到原料限制。为构建高效的微生物细胞工厂,采用合成生物学手段,利用模块组装方法将啤酒花来源的关键结构基因整合到同一载体上,并以组成型启动子进行表达,在酿酒酵母中实现蛇麻酮异源合成。此外,敲除了酿酒酵母内源亮氨酸分解关键基因,进一步提高了蛇麻酮类化合物产量,为研究其他苦味酸化合物的合成机制奠定了基础。
Lupulone homologs,the prenylated phloroglucinol derivatives,contribute the flavor of beer and have bioactivities of anti-microbial and anti-cancer.Lupulone homologs are mainly separated and purified from hops,which is limited by the cost and the resources of raw materials.To construct the high-efficient microbial factory,synthetic biology tools were taken to integrate the heterogenous genes from hops into one single plasmid with modular pathway engineering strategy,and the genes were expressed with constitutive promoters,realizing the heterogenous synthesis of lupulone homologs in Saccharomyces Cerevisiae.Besides,the deletion of the key gene in the degradation of leucine further improved the yields of lupulone homologs.This work provides an approach for the further research of the synthetic mechanism of other bitter acids in yeast.
