斑点叉尾鮰维氏气单胞菌的分离鉴定及其病理损伤研究

摘要
为确定一起斑点叉尾鮰体表褪色及胃肠道炎症的病原,为斑点叉尾鮰肠炎的正确诊断及防治提供理论依据。从病鱼脾脏和肾脏分离到两株优势菌(DYJ0320-1、DYJ0320-2),检测分离菌的形态、生理生化特性,以腹腔注射的方式进行人工感染试验,同时对16S rRNA和gyrB基因序列测定(Genebank登录号分别为KU925194和KU925195)与系统发育分析,并进行药敏试验。两株菌的生理生化特性均与已报道的维氏气单胞菌(Aeromonas veronii)相一致;16S rRNA和gyrB基因序列测定将其鉴定为维氏气单胞菌。病鱼多个组织器官具有典型的病理变化,分别引起坏死性、出血性胃肠炎,出血性脾炎及间质性肾炎,坏死性、溃疡性皮炎。药敏结果显示该菌对恩诺沙星、头孢西丁及左氧氟沙星等药物敏感。导致斑点叉尾鮰本次体表褪色及胃肠道炎症的主要病原菌为维氏气单胞菌。
To identify the pathogens caused the faded skin and inflammation of the digestive tract in Channel catfish, and provide a theoretical basis on correct diagnosis and prevention of channel catfish enteritis. Two dominant bacteria(DYJ0320-1、DYJ0320-2)had been isolated from the kidney and spleen of diseased fish. They were identified as Pseudomonas putida based on morphology, biochemical, physiology characteristics and sequencing of 16 S rRNA and gyrase B subunit(gyrB) gene(Gene Bank accession number were KP693689 and KP693690 respectively).They were also proved to be the pathogen of the disease by artificial infection experiment.Histologically, Damaged organs characterized by hemorrhagic necrotizing inflammation of the digestive tract, hemorrhagic inflammation of the spleen and interstitial nephritis, necrotizing, and ulcerative dermatitis. The result of drug sensitivity test showed that the isolates were senitive to the Enrofloxacin, Cefoxitin, Azithromycin, Levofloxacin, but were resistant to Ampiciuin,Doxycycline, novobiocin, Amoxicillin, Rifarnpin. The isolates proved to be the pathogen of channel catfish enteritis and was identified as Aeromonas veronii.
