冰温贮藏过程中罗非鱼片蛋白质表达谱分析

摘要
本文采用了label free定量法鉴定了冰温贮藏过程中0天和7天罗非鱼片的蛋白质表达谱。一共鉴定出了902种蛋白质,其中154种为差异表达蛋白,294种缺失蛋白。对全部蛋白进行GO功能注释和KEGG代谢通路分析发现,生物过程主要为细胞过程,代谢通路则为主要的KEGG通路。差异表达蛋白的主要分子功能为结合以及催化活性功能。缺失的蛋白质主要涉及代谢过程,氧化还原过程,翻译和运输等。该研究对罗非鱼骨骼肌蛋白质组与鱼肉鲜度和品质的关系有显著的贡献,并为鱼肉鲜度变化机制及功能研究提供了新的视角。
Protein profile of the skeletal muscle of tilapia, Oreochromis Niloticus, was carried out with label-free quantitative proteomic analysis. A total of 902 tilapia muscle proteins were identified, and 154 differentially expressed proteins and 294 absent proteins from the pool of all experiments run in this study. Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG) were carried out for functional annotation and for pathway annotation. The analysis of total proteins differentially expressed proteins revealed that the top-ranking biological process was cellular process; and metabolic pathway was the top-ranking KEGG pathway. The differentially expressed proteins were focused on binding and catalytic activity in molecular function. The major biological processes of absent proteins were metabolic process, oxidation-reduction process, translation and transport. This work makes a significant contribution towards the freshness and quality of tilapia skeletal muscle proteome and provides novel insights into its role in freshness mechanism and function.
