纤维素酶法破壁条件对雨生红球藻类胡萝卜素提取效果的影响

摘要
为了评价纤维素酶法破壁在雨生红球藻类胡萝卜素提取中的应用价值,研究了酶浓度、酶反应pH、破壁温度和破壁时间4个因素对破壁后雨生红球藻类胡萝卜素提取的影响,并对破壁条件进行了优化。试验结果表明:酶浓度、酶反应pH、破壁温度和破壁时间都会对雨生红球藻破壁提取类胡萝卜素产生显著影响,但以破壁温度及酶浓度对类胡萝卜素提取量影响最大。用正交试验法得到优化的纤维素酶法破壁雨生红球藻的条件为:酶浓度0.6mg/mL、酶反应pH 4.0、破壁温度50℃、破壁时间30 min,优化后提取物中总类胡萝卜素的含量为59.62±0.31mg/g DW,比优化前提高了25%。
In order to evaluate the possibility of extracting carotenoid from Haematococcus pluvialis disrupting cell wallby cellulase,influences of several factors such as enzyme concentration,enzyme reaction pH,disrupting temperature and disrupting time on extracting carotenoid from Haematococcus pluvialis were studied and conditions used to disrupt the cell wall were optimized.Results indicated that factors such as enzyme concentration,enzyme reaction pH,disrupting temperature and disrupting time had significant effects on the extracting content of carotenoid,and disrupting temperature and enzyme concentration were the most significant for the extracting content compared with the other factors.The optimal conditions gained by orthogonal experiment were:enzyme concentration 0.6 mg/mL,enzyme reaction pH 4.0,disrupting temperature 50 ℃,disrupting time30 min.Under the optimized condition,thetotal carotenoid content in the extracting liquid was 59.62 ±0.31 mg/g DW,which increased 25%comparing with that of the non-optimized condition.
