空间诱变野鸦椿组培快繁技术研究

摘要
以空间诱变野鸦椿带芽茎段为材料,进行组织培养快速繁殖研究。结果表明:最佳消毒方式为采摘后室内放置5d再进行消毒,以75%酒精消毒30s结合0.1%升汞溶液消毒8min效果最佳,污染率与死亡率均处于相对较低水平;适宜的初代培养基为MS+1.0mg·L~(-1)+NAA 0.1mg·L~(-1),萌芽率为94.44%;适宜的增殖培养基为MS+1.0mg·L~(-1)+NAA 0.1mg·L~(-1),增殖系数为4.12;适宜的生根培养基为1/3WPM+IBA0.3mg·L~(-1)+NAA0.3mg·L~(-1),生根率为69.23%。
Stem segments with buds were used as explants to study tissue culture and rapid propagation of Euscaphis japonica(Thunb.) Kanitz,which seeds had been through space mutation.The results indicated that the explants in the room for 5d after 75%alcohol disinfection 30 s and then in 0.1%HgCl_2 disinfection 8 min is suitable,the contamination rate and mortality rate are in lower level.The suitable culture medium for primary culture was MS + 1.0mg · L~(-1)+ NAA 0.1mg · L~(-1) with the rate of effective germination of 94.44%.The suitable culture medium for subculture was MS + 1.0mg · L~(-1) + NAA 0.1mg ·L~(-1) with the proliferation value of 4.12.The suitable culture medium for rooting was 1/3WPM +IBA 0.3 mg · L~(-1)+ NAA0.3mg · L~(-1),the rate of rooting was 69.23%.
