近红外响应纳米材料在肿瘤影像示踪与诊疗一体化中的应用研究

摘要
癌症是威胁人类生命的一种重大疾病。目前,癌症的治疗方法主要有化疗、放疗和手术治疗,但是这些治疗手段在临床使用中遇到许多困难,使得癌症的临床治疗效果并不理想。然而,纳米技术的快速发展为癌症诊疗带来了新的希望。近红外响应纳米粒子因其具有尺寸较小、光热转换效率高以及X-射线吸收能力强等特点在癌症探测、诊断和治疗领域有广泛应用。我们通过优化近红外响应纳米材料构建稳定、高效和安全的纳米载体,并将化疗、光动力学治疗等治疗方式相结合,发展整合靶向、活体示踪、光热治疗、药物治疗和预后检测等功能于一体的诊断治疗纳米药物。我们的结果表明,近红外响应纳米材料具有良好的生物相容性,其自身可作为优良的光声成像造影剂、CT造影剂以及光热试剂;同时利用其两亲的表面负载抗肿瘤药物或者光敏剂,可以实现药物的可控缓释。这将为有效地提高纳米药物呈递效率和减轻药物毒副作用提供强有力的支持。
Cancer is one of the greatest killers and threats to human life in the worldwide. Nowadays, as each treatment method such as chemotherapy has its drawbacacks, cancer therapy still doesn't reach an optimal result. However, the rapid development of nanotechnology brights about new opportunities for cancer diagnosis and therapeutics. Recently, due to the small size, high NIR absorption coefficient and X-ray attenuation capability, NIR-responsive nanomaterials have been used for cancer detection, diagnosis and treatment widely. Herein, we mainly focus on building stable, efficient and safe nanocarriers, ultimately fabricating theranostic agents integrating targeted therapy, in vivo tracing and prognosis monitoring into one system. Our results indicate that these theranostic agents possess ideal photothermal effect and contrast enhancement in MSOT/CT bimodal imaging, while being developed as nanocarriers for NIR photothermal-triggered drug delivery. This simple but powerful theranostic nanomaterial shows a potential promise for precision nanomedicine paradigm.
