植物乳杆菌二元信号系统的结构分析与功能预测

摘要
二元信号系统(TCS)是原核生物中广泛存在的一种应激响应系统,由组氨酸蛋白激酶(HK)和反应调节器(RR)组成。本研究利用NCBI结合原核TCS查询系统,针对植物乳杆菌(L.plantarum)的6个菌株(WCFS1,P-8,JDM1,16,ZJ316和ST-Ⅲ)进行了TCS查找、结构分析和功能预测。结果发现,L.plantarum含有13个典型的TCS,1个HK"孤子"和2个RR"孤子"。大多数HKs/RRs含有5个以上跨膜区域,可能感应胞膜信号;TCSs HK1/RR1、HK4/RR4、HK7/RR7、HK12/RR12、LamC/LamA、PlnB2/PlnC/PlnD存在较大胞外区域,可能感应胞外信号。TCSs可能涉及脂质合成、低氧调控、群体感应、黏附能力、生物膜形成、代谢酶合成以及细菌素产生等多项功能。该研究有助于全面系统地了解L.plantarum TCS的结构,并为其功能解析、调控机制和调控靶点的研究奠定基础,也能为食品工业应用的L.plantarum基因工程改良提供参考。
Two-component signal transduction systems(TCSs) are stress response systems widely found in prokaryotes,and consist of a histidine kinase(HK) and a response regulator(RR).In this paper,the TCSs in 6 Lactobacillus plantarum(L.plantarum) strains(WCFS1,P-8,JDM1,16,ZJ316,and ST-Ⅲ) were searched by using NCBI and the inquiry system of prokaryotic TCSs,and their structures and functions were analyzed and predicted.Thirteen TCSs,1 HK ' Orphan' and 2 RR'Orphans' were found in these L.plantarum strains.Some HKs-RRs contain 5 or more transmembrane regions,which could sense the signal on cytoplasma membrane.We found that the HK3 and HK11 of the sensing element of TCSs may be induced by intracellular signals.Each of the other TCSs,including HK1-RR1,HK2-RR2,HK4-RR4,HK5-RR5,HK7-RR7,and HK12-RR12,has a large extracellular region that could possibly sense the signal outside the cell.The TCSs are probably involved in such functions as lipid synthesis,hypoxia regulation,quorum sensing,adhesion ability,biofilm formation,metabolic enzyme synthesis,and bacteriocin production.This study contributes to the comprehensive and systematic understanding of the structure of TCSs of L.plantarum strains,and could provide a basis for the study of TCS functions,regulating mechanisms,and targets.Moreover,it can serve as a reference for genetic engineering improvement of L.plantarum used in food industry.
