rGO-SnO_2复合材料:一种双功能亲和探针对于内源性多肽和磷酸化多肽的高效富集

低丰度内源性多肽和磷酸化多肽在生理病理学过程中扮演着重要角色~1,体内这两种多肽含量的变化与疾病的发生发展息息相关,因此,对两者的研究在临床检测和生物标志物的发现上有重要意义。然而,内源性多肽的检测受基质干扰很大。另一方面,磷酸化多肽由于丰度低,离子化困难,受其他非磷酸化多肽信号抑制严重而难以用质谱进行直接检测~2。因此,在质谱分析之前,对样品进行选择性富集尤为重要。本工作通过一步水热反应法合成氧化石墨烯-二氧化锡复合物,再通过对其热处理得到还原氧化石墨烯-二氧化锡复合材料rGO-SnO_2。结合rGO的疏水性和SnO_2的高亲和性,该材料可实现对低丰度多肽和磷酸化多肽的分布富集。rGO-SnO_2总共能从BSA酶解液中富集到34条多肽,对磷酸化多肽的检出限达到4×10~(-10)M,成功实现对复杂生物样本人血清中的内源性多肽和磷酸化多肽的分布富集。该结果展示了复合材料的双功能作用,给样品前处理领域的材料设计提供了新的思路。
Low-abundance endogenous peptides and phosphopeptides play crucial roles in physiological and pathological processes.Changes in these two peptides are closely related to various disease initiation and progression,thus related research is important for clinical diagnostic and biomarker discovery.However,the analysis of endogenous peptides suffer from complex matrix and the low stoichiometry,poor ionization,and signal suppression make it difficult to analyse phosphorylation by mass spectrometry(MS) directly.Powerful sample preparation prior MS analysis is in indispensable.Herein,we synthesized composite rGO-SnO_2 through one-pot solventhermal process,followed by thermal reduction.With the hydrophobicity of rGO and high affinity of SnO_2,rGO-SnO_2 was used for sequential enrichment of endogenous peptides and phosphopeptides.As a result,34 peptides were captured from BSA digest and the LOD of phosphopeptides from β-casein digest was 4×10~(-10) M,the material showed feasibility in human serum.This multifunctional affinity probe opens a new perspectives in material design in sample preparation.
