酰肼修饰单分散二氧化硅微球用于磷酸化肽的灵活可控富集

摘要
在本工作中,我们设计并制备了一种酰肼修饰的二氧化硅单分散微球,并将这种材料应用于对于包含不同磷酸化位点数目的磷酸化肽的可控选择性富集。所制备的材料具有规整的球形形貌,其直径约200 nm并具有很好的单分散性,材料表面所修饰的酰肼基含量约为0.5 mmol/g。该材料对于包含不同磷酸化位点数目的磷酸化肽的吸附和解吸附可以通过简单调节缓冲液中甲酸的浓度来进行灵活的控制。当缓冲液中含有0.02%的甲酸时,材料可以同时吸附单磷酸化肽和多磷酸化肽;当缓冲液中含有0.5%的甲酸时,材料可以吸附多磷酸化肽而不吸附单磷酸化肽;而当甲酸浓度进一步提高到2%时,材料则仅吸附包含三个以上磷酸化位点的多磷酸化肽。借助于材料这种对包含不同磷酸化位点数目磷酸化肽的灵活可控的吸附性质,我们通过不同的上样缓冲液和洗脱缓冲液的组合,成功实现了对于标准磷酸化蛋白样品和复杂实际样品中磷酸化肽的全富集分析以及对包含不同磷酸化位点数目的磷酸化肽的分别富集分析。
In this work,we designed and synthesized hydrazide modified silica monodispersed microsphere for tunable and versatile enrichment of phosphopeptides with different numbers of phosphorylation sites.This material was characterized by its uniform spherical morphology and diameter of ca.200 nm with good monodispersity,and 0.5mmol/g of hydrazide groups were loaded on the surface of this microsphere.Adsorption of phosphopeptides with different numbers of phosphorylation site can be adjusted by simply changing the concentration of formic acid in buffers.Both mono- and multi- phosphorylated peptides can be adsorbed in buffers with lower concentration of formic acid,while only phosphopeptides with more phosphorylation sites were preferred in buffers with higher concentration of formic acid.By choosing different combination of loading buffer and elution buffer,both simultaneous enrichment of all phosphopeptides and separate enrichment of phosphopeptides with different numbers of phosphorylation sites were realized in standard phosphoprotein and real sample digest.
