重金属离子神经毒性的细胞代谢组学研究

<正>重金属离子铜、铅和镉是有毒的环境和工业污染物,它们能够透过血脑屏障进入人脑并在脑内蓄积,极性和慢性暴露会导致神经毒性,从而引起各种脑、神经系统等疾病[1]。目前,有关它们神经毒性的报道很多,但是关于它们毒性作用的分子机制研究较少。我们运用细胞代谢组学的研究方法,以LC-MS为分析测
The heavy metals Cd~(2+),Pb ~(2+)or Cd~(2+) are toxic environmental and industrial pollutants.Acute or chronic exposure can result in neurotoxicity.However,the neurotoxicity mechanisms of these metals are still not clear.To explore the underlying mechanism,the neuronal PC-12 cells were used and treated with CuSO_4,PbCl_2 and CdCl_2.And the intracellular and extracellular metabolites were profiled using ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry.These heavy metals could give rise to excessive oxidative stress to lead to cell death.The metabolic changes were involved in phenylalanine and tryptophan metabolism,pantothenate and CoA biosynthesis,Pyrimidine metabolism,protein biosynthesis and the others.These results provide insight in the mechanism of neurotoxic research of heavy metals.
