液相色谱-质谱法分析皱纹盘鲍性腺中的磷脂

皱纹盘鲍因口感好、营养丰富,受到科研工作者和消费者的广泛关注,但在加工过程中,约20%的内脏、性腺等副产物未被充分利用。皱纹盘鲍性腺富含油脂,且磷脂含量丰富。因此,本文以雌、雄皱纹盘鲍性腺为原料,以磷脂酰胆碱和磷脂酰乙醇胺为研究对象,采用液相色谱质谱法分析了鲍鱼性腺中磷脂的结构。本文首先采用MTBE法提取了皱纹盘鲍性腺中的总脂,雌性皱纹盘鲍性腺组油脂提取率为34.44±1.77%,雄性性腺组油脂提取率为15.03±1.22%。然后采用棒状薄层色谱法分析了脂质组成,雌性皱纹盘鲍性腺组磷脂相对含量为59.60±5.96%,雄性性腺组磷脂相对含量为47.84±6.92%。最后采用液相色谱质谱法对皱纹盘鲍性腺中的磷脂酰胆碱和磷脂酰乙醇胺进行了定性和定量分析,并对分子种进行了鉴定。结果表明,雌性皱纹盘鲍性腺中磷脂酰乙醇胺种类及含量高于雄性组,雄性鲍鱼性腺中磷脂酰胆碱的种类和含量则高于雌性性腺组。
Abalone(haliotis discus hannai ino) has captured increasing interest of scientists and consumers due to its good taste and rich nutrition.However,almost 20%of by-products such as the guts and gonads are underutilized during processing.The gonads are rich in lipids,in particular phospholipids(PL).Hencethis current study was carried out to identify the phosphocholine(PC) and phosphoethanolamine(PE) compositions of lipids extracted from female and male abalone gonads by using HPLC-ESI-MS/MS.In this study,the total lipid extraction method of MTBE was employed to recover lipids from female and male abalone gonads,and the oil extraction rates of these two kinds of abalone gonads were 34.44 ± 1.77%and 15.03 ± 1.22%,respectively.Then the lipid class compositions were determined by using thin layer chromatography-flame ionization detection(TLCFID).Results indicated that PL accounted for 59.60 ±5.96%and 47.84 ±6.92%,respectively,of the total lipid for female and male abalone gonads.Besides,the molecular species and relative content of PC and PE were analysed by using HPLC-ESIMS/MS.Results indicated that the content and species of PC in female abalone gonads were higher than male's,while the content and species of PE in male abalone gonads were higher than female's.
