不同原料和溶剂制备的南极磷虾油中磷脂酰胆碱和磷脂酰乙醇胺的组成分析

本研究首先采用Folch法从不同原料(南极磷虾全虾、虾壳、虾肉冻干粉)中提取南极磷虾总脂,然后以南极磷虾冻干全虾粉为原料,以乙醇、正己烷、乙醇:正己烷(1:2,v/v)为溶剂提取南极磷虾油。通过液相色谱串联质谱法(HPLC-ESI-MS/MS)对上述样品中的磷脂酰胆碱(GPCho)和磷脂酰乙醇胺(GPEtn)进行定性和内标法半定量分析,并对分子种进行鉴定。研究结果表明,南极磷虾总脂中共含有55种GPCho和32种GPEtn。其中,磷脂酰脂(Phosphatidyl)16:0/18:1、16:0/20:5、16:0/22:6为GPCho的主要存在形式,磷脂酰脂(Phosphatidyl)16:0/18:1、16:0/20:5、16:0/22:6、18:1/20:5为GPEtn的主要存在形式。根据含有二十碳五烯酸(EPA)和二十二碳六烯酸(DHA)的GPCho和GPEtn的相对含量可知,虾壳适合做GPCho的浸提原料,全虾适合做GPEtn的浸提原料,而乙醇:正己烷(1:2,v/v)适合做GPCho的浸提溶剂,正己烷适合做GPEtn的浸提溶剂。
In this study,total lipids in raw materials(whole krill powder,krill meat powder and krill shell powder) were extracted by a modified method of Folch,krill oils(KO) were extracted from whole krill powder by using the organic solvents(ethanol,hexane,and a mixture of ethanol and hexane(1:2,v/v)) method.Then using the method of highperformance liquid chromatography-electrospray ionization-tandem mass spectrometry(HPLC-ESI-MS/MS) to detect the separation,identification and semi-quantification of glycerophosphocholine(GPCho) and glycerophosphoethanolamine(GPEtn) molecular species in samples.The results showed that at least 55 GPCho species and 32 GPEtn species were observed in total lipids from materials of different tissues including whole krill,krill shell and krill meat.Phosphatidyl GPCho 16:0/18:1,16:0/20:5,and 16:0/22:6,and phosphatidyl GPEtn 16:0/18:1,16:0/20:5,16:0/22:6 and18:1/20:5 were the major molecular species.According to the amounts of molecular species containing EPA and DHA,krill shell is the most suitable raw material for GPCho,while whole krill is the most suitable raw material for GPEtn,and the mixture of ethanol and hexane(1:2,v/v) is the most suitable solvent for GPCho,but hexane is the most suitable solvent for GPEtn.
