痛风康宁方对急性痛风性关节炎大鼠血清炎性因子及氧化应激的影响

摘要
目的:通过观察痛风康宁方对痛风性关节炎大鼠血清炎性因子及氧化应激的影响,探讨其初步的作用机制。方法:48只Wistar雄性大鼠随机分为对照组,模型组,秋水仙碱组,痛风康宁低、中、高剂量组,每组8只。除对照组,余各组采取黄嘌呤(XA)灌胃联合腹腔注射氧嗪酸钾(OAPS),连续7d,同时各药物组给予药物干预,并于造模后5d向踝关节腔注射尿酸钠建立急性痛风性关节炎模型,之后48h终止实验。分别于踝关节腔注射尿酸钠前后测定其踝关节肿胀程度,实验终止时检测血清尿酸(UA)、黄嘌呤氧化酶(XOD)、白介素1-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)、丙二醛(MDA)及一氧化氮(NO)含量,观察大鼠踝关节病理组织学变化。结果:与对照组比较,模型组踝关节肿胀度明显(P<0.05),UA、XOD、IL-1β、TNF-α、MDA及NO水平均显著升高(P<0.01),SOD含量明显减少(P<0.01);病理学显示,模型组踝关节滑膜组织大量炎性细胞浸润,细胞增生明显;与模型组比较,痛风康宁低、中、高剂量组,均可以显著降低大鼠踝关节肿胀度(P<0.05),大鼠UA、XOD、IL-1β、TNF-α、MDA及NO水平均明显降低(P<0.01,P<0.05),SOD含量显著增加(P<0.01),关节组织炎性浸润,细胞组织增生程度明显减轻。结论:痛风康宁方对急性痛风性关节炎有显著的抗炎作用,其作用机制与抑制XOD活性,降低血UA水平,降低IL-1β、TNF-α、MDA、NO的表达,促进SOD的释放有关。
        Objective: To investigate the effects of Tongfeng Kangning Formula on serum inflammatory factors and oxidative stress in rats with gouty arthritis. Methods: Forty-eight Wistar male rats were randomly divided into control group, model group, colchicine group, and Tongfeng Kangning Formula low, medium and high dose groups, with 8 rats in each group. In addition to the control group, the remaining groups were treated with xanthine(XA) and intraperitoneal injection of potassium oxonate(OAPS) for 7 days. At the same time, each drug group was given drug intervention and injected into the ankle joint cavity 5 days after modeling. Sodium urate was used to establish an acute gouty arthritis model, and the experiment was terminated 48 h later. The degree of swelling of the ankle joint was measured before and after injection of sodium urate in the ankle joint cavity. Serum uric acid(UA), superoxide dismutase(XOD), interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α), superoxide dismutase(SOD), malondialdehyde(MDA) and nitric oxide(NO) were detected at the end of the experiment. The histopathological changes of ankle joint in rats were observed. Results: Compared with the control group, the degree of swelling of the ankle joint in the model group was significantly(P<0.05), and the levels of UA, XOD, IL-1β, TNF-α, MDA and NO were significantly increased(P<0.01),and the SOD content was significantly reduced.(P<0.01); pathology showed that a large number of inflammatory cells infiltrated in the synovial tissue of the model group, and cell proliferation was obvious; compared with the model group, the lower, middle and high dose groups of Tongfeng Kangning Formula could reduce the degree of joint swelling(P<0.05), the levels of UA, XOD, IL-1β, TNF-α, MDA and NO in rats were significantly decreased(P<0.01, P<0.05), and the content of SOD was significantly increased(P<0.01). Inflammatory infiltration of joint tissue, the degree of cell tissue proliferation was significantly reduced. Conclusion: Tongfeng Kangning Formula has significant anti-inflammatory effects on acute gouty arthritis. Its mechanism is related to inhibiting XOD activity,lowering blood uric acid level, decreasing the expression of IL-1β, TNF-α, MDA and NO, and promoting the release of SOD.
