HPLC法同时测定荜茇三味散中胡椒碱和荜茇宁的含量

摘要
目的:建立测定由山楂、诃子和荜茇组成的蒙药荜茇三味散中胡椒碱和荜茇宁含量的HPLC方法。方法:在室温下,使用Waters Symmetry C18(250mm×4.6mm,5.0!m)柱;采用水-乙腈(50∶50)为流动相,流速1mL·min~(-1),进样量为10μL;检测波长343nm。结果:荜茇宁和胡椒碱分别在0.25～12.50μg·mL~(-1)(r=0.999 0)和400～800μg·mL~(-1)(r=0.999 0)浓度范围内与峰面积呈良好的线性关系,平均回收率分别为100.63%和97.09%(n=5)。荜茇三味散中胡椒碱和荜茇宁含量分别为42.014μg·mg~(-1)和458.76μg·mg~(-1)。结论:该方法简便、精准、回收率高,可用于蒙药荜茇三味散中胡椒碱和荜茇宁含量的同时测定。
Objective:To establish a method for simultaneous determination of piperine and piperlonguminine in Mongolian medicine Biba-3 Wei San by HPLC with Waters Symmetry C_(18)(250 mm×4.6 mm,5.0μm)column.Methods:Under the conditions of the room temperature,acetonitrile and water(50∶50)as the mobile phase,the flow rate 1 mL/min and the injection volume 10μL,the analytes were detected at 340 nm.Results:The calibration curves showed good linearity in the concentration ranges of piperlonguminine 0.25～12.50μg·mL~(-1)(r=0.999 0)and piperine 400～800μg/mL(r=0.999).The average recoveries have been found to be100.63% and 97.07%,and the contents were 42.014μg·mg~(-1) and 458.76μg·mg~(-1),respectively.Conclusion:This method is simple,accurate and high recoveries,which can be applied for monitoring the contents of piperine and piperlonguminine in Mongolian medicine.
