葛根芩连汤对2型糖尿病大鼠胰岛素抵抗及炎性因子的影响

摘要
目的通过观察葛根芩连汤对2型糖尿病(T2DM)大鼠胰岛素抵抗及炎性因子的影响,探讨其改善胰岛素抵抗的作用机制。方法从40只大鼠中随机选取10只为空白组,余采用高脂高糖饮食饲养方法加注射小剂量链脲佐菌素(STZ)制备T2DM模型。将模型制备成功的大鼠随机分为模型组、罗格列酮组和葛根芩连汤组。按10 mL/kg给予空白组和模型组大鼠0.9%氯化钠溶液,同时罗格列酮组和葛根芩连汤组分别被给予罗格列酮溶液3 mg/kg和葛根芩连汤18.2 g/kg灌胃,1次/d,共4周。运用血糖仪检测各组大鼠FBG,以及放射免疫法检测FINS水平,并计算HOMA-IR;采用ELISA检测各组大鼠血清炎性因子TNF-α、IL-6、IL-1β以及MCP-1的水平。结果与模型组比较,罗格列酮组大鼠的FBG显著下降(P <0.01);与模型组比较,罗格列酮组和葛根芩连汤组大鼠的FINS、HOMA-IR及血清TNF-α、IL-6、IL-1β、MCP-1水平明显降低(P <0.01或P <0.05)。结论葛根芩连汤可改善T2DM大鼠胰岛素抵抗,该作用可能与抑制炎性因子释放有关。
        Objective To observe the effect of Gegen Qinlian Decoction on insulin resistance and inflammatory factors in type 2 diabetes mellitus(T2 DM) rats and explore the mechanism of ef fect. Methods Ten rats were randomly selected from 40 T2 DM rats as blank group, and other 30 rats were fed with high fat and sugar diet and injected lowdose streptozotocin(STZ) to prepare the model of T2 DM. The successful T2 DM model rats were randomly divided into model group, rosiglitazone group and Gegen Qinlian Decoction group. The rats of blank group and model group were given 0.9% sodium chloride solution(10 mL/kg), moreover the rats of rosiglitazone group and Gegen Qinlian Decoction group were given rosiglitazone solution 3 mg/kg and Gegen Qinlian Decoction 18.2 g/kg respectively, once a day for 4 weeks. FBG was measured by blood glucose meter, and FINS was measured by radioimmunoassay and then HOMA-IR was calculated. The levels of serum inflammatory factors, including TNF-α, IL-6, IL-1β and MCP1,were detected by ELISA. Results Compared with the model group, FBG in rosiglitazone group was significantly decreased(P < 0.01), but there were no significant difference between Gegen Qinlian Decoction group and model group rosiglitazone group. Compared with model group, the levels of FINS, HOMA-IR and serum TNF-α, IL-6,IL1β and MCP-1 in rosiglitazone group and Gegen Qinlian Decoction group were significantly decreased(P < 0.01 or P < 0.05). Conclusion Gegen Qinlian Decoction could improve insulin resistance in T2 DM rats, which might be related to inhibit the release of inflammatory factors.
