摘要
目的:采用高效液相-紫外检测器色谱法测定低聚木糖含量。方法:利用1-苯基-3-甲基-5-吡唑啉酮(PMP)进行衍生,采用高效液相-紫外检测器色谱仪测定低聚木糖含量。结果:色谱柱:ACE C18(4.6mm×250mm);流动相:乙腈-0.02mol/L乙酸铵溶液(17∶83);柱温:30℃;波长:250nm;流速:1.0mL/min;进样量:5μL。低聚木糖在0.02~0.2mg·mL~(-1)浓度范围内与峰面积积分呈良好的线性关系,回归方程为y=1.1281x+0.0123,R~2=0.9996。结论:建立的方法准确可靠,重复性好,分离度高,灵敏度强且操作简单,可用于测定低聚木糖的含量。
Objective:To establish Testing method and Methodological investigation of xylo-oligosaccharide by HPLC-UV method.Methods:The content of xylo-oligosaccharide was determined by HPLC using derivatization of 1-phenyl-3-methyl-5-pyrazolinone(PMP).Results:Chromatographic conditions:chromatographic column:ACE C18(4.6 mm×250 mm),Mobile phase:ammonium acetate solution of acetonitrile-0.02 mol/L(17∶83),Column temperature:30℃;Wavelength:250 nm,Flow rate:1.0 mL/min,Sampling quantity:5μL.In the range of 0.02~0.2 mg·mL~(-1),the peak area has a good linear relationship with the sample intake.The regression equation is y=1.1281 x+0.0123,R~2=0.9996.Conclusion:The method is accurate and reliable,and of good repeatability,high separating degree and sensitivity,and it is simple and easy to implement.It can be used to determine the content of oligosaccharide.
