摘要
目的:探讨外源性十溴联苯醚(BDE-209)暴露对小鼠早期胚胎发育的影响。方法:选择健康昆明雌性小鼠40只、雄性小鼠10只。40只雌小鼠随机分成4组:BDE-209低剂量组(100 mg/kg)、BDE-209中剂量组(300 mg/kg)、BDE-209高剂量组(500mg/kg)和对照组,每组10只。所有雌小鼠采用灌胃方式进行BDE-209染毒,对照组给予等量生理盐水。观察比较各组体外受精(IVF)情况及卵泡液中肿瘤坏死因子-α(TNF-α)和干扰素-γ(IFN-γ)浓度。结果:BDE-209中剂量组及高剂量组受精率明显低于对照组(P<0.05),但BDE-209低剂量组与对照组比较差异无统计学意义(P>0.05)。与对照组相比,BDE-209低、中、高剂量组4-细胞、8-细胞、桑椹胚及囊胚形成率均明显下降,卵泡液中TNF-α及IFN-γ浓度均明显升高(P<0.05),且随着BDE-209暴露剂量的增加呈降低/升高趋势,不同剂量组间比较差异有统计学意义(P<0.05)。结论:外源性BDE-209暴露会减少小鼠IVF的获卵率,降低受精率,对小鼠早期胚胎发育产生负性影响,且呈剂量依赖性。
        Objective:To investigate the effect of exogenous BDE-209 exposure on early embryonic development in mice.Methods:40 female mice were randomly divided into 4 groups:low-dose(100 mg/kg),middle-dose(300 mg/kg),and high-dose(500 mg/kg)BDE-209 groups and control group,with 10 mice in each group.Mice were administered different doses of BDE-209 by gavage except for the controls(received equal volume of saline).The in vivo fertilization(IVF)status,and the levels of tumor necrosis factor(TNF)-αand interferon(IFN)-γin follicular fluid were observed and compared.Results:The fertilization rate in highdose and middle-dose BDE-209 groups was significantly lower than that in the control group(P<0.05),while no significant difference was found between the low-dose group and the control group(P >0.05).Compared with the control group,the 4-to 8-cell embryos and blastocyst formation rate were reduced,and while the levels of TNF-αand IFN-γin follicular fluid were elevated in the BDE-209-treated groups(P<0.05).Conclusion:BDE-209 exposure could result in decreased fertility rate and blastocyst formation rate in mice,and was dose-dependent.
