摘要
目的:建立HPLC法同时测定凌霄花中齐墩果酸和熊果酸,比较不同产地的凌霄花中两种成分的含量。方法:选用Shim-pack VP-ODS C_(18)(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.1%磷酸为流动相进行梯度洗脱,流速为1.0 mL/min,检测波长为215 nm,柱温为30℃。结果:齐墩果酸和熊果酸进样量分别在0.233 2~5.830 0μg和0.274 8~6.870 0μg范围内呈线性,回归方程分别为Y=6.337×10~5X+1.538×10~3(r=0.999 6)和Y=8.158×10~5X+2.977×10~3(r=0.999 7),平均加样回收率依次为98.9%和98.7%。采自连云港、南京的凌霄花中齐墩果酸和熊果酸的含量略高于其他4个产地。结论:所建立的方法可以准确的对凌霄花中齐墩果酸和熊果酸进行含量测定,不同产地的凌霄花中两种成分的含量存在差异。
        Objective: To establish the HPLC method for simultaneous determination of oleanolic acid and ursolic acid in Chinese trumpet vine, and compare the content of two components in Chinese trumpet vine from different habitats.Methods: Selected Shim-pack VP-ODS C_(18)(250 mm × 4.6 mm, 5 μm) chromatographic column, and conducted a gradient elution with acetonitrile-0.1% phosphoric acid as mobile phase,with flow rate being 1.0 mL/min, detection wavelength 215 nm, and column temperature 30 ℃. Results: The values of oleanolic acid and ursolic acid were linear in the range of 0.233 2~5.830 0 μg and 0.274 8~6.870 0 μg,respectively. The regression equations were Y=6.337 × 10~5 X+ 1.538×10~3(r=0.999 6) and Y=8.158×10~5 X+2.977×10~3(r=0.999 7),and the average sample recovery was 98.9% and 98.7% respectively. The content of oleanolic acid and ursolic acid in Chinese trumpet vine from Lianyungang and Nanjing was slightly higher than that in the other four habitats. Conclusion: The established method can be used to determine the content of oleanolic acid and ursolic acid in Chinese trumpet vine,and there were differences in the content of two components in different habitats of Chinese trumpet vine.
