浙贝母-知母药对的止咳、化痰及平喘作用研究

摘要
目的:考察浙贝母-知母药对配伍的止咳、化痰及平喘作用并探讨其机制。方法:分别采用小鼠氨水引咳法和豚鼠枸橼酸引咳法研究药对的止咳作用;ELISA法测定小鼠血清中白细胞介素-10(IL-10)水平,观察其止咳机制;小鼠气管酚红排泌法研究化痰作用;豚鼠整体动物引喘法研究平喘作用。结果:与相应的空白对照组比较,浙贝母-知母药对配伍中、高剂量可明显延长小鼠和豚鼠的咳嗽潜伏期并减少咳嗽次数(P<0.05);增加小鼠血清IL-10的释放(P<0.05);显著增加小鼠呼吸道黏膜酚红的排泌量(P<0.05);显著延长豚鼠哮喘潜伏期(P<0.05)。结论:浙贝母-知母配伍具有明显的止咳、化痰及平喘作用,其止咳机制可能通过外周抗炎作用实现。
Objective: To observe the effects of couplet medicines Fritillaria thunbergii and Rhizoma anemarrhenae on relieving cough, eliminating phlegm and relieving asthma and discuss its mechanism. Methods: The cough relieving effect of couplet medicines were studied using ammonia water-induced tussive mice and citric acid-induced tussive guinea pig. The mechanism of relieving cough were studied by measuring the levels of interleukin-10(IL-10) in serum by enzyme-linked immunosorbent assay(ELISA) method. The phlegm elimination effect were researched by the observation of tracheal phenol red shedding in mice. The asthma relieving effect were tested by spraying method in guinea pig. Results: The couplet medicines Fritillaria thunbergii and Rhizoma anemarrhenae could significantly extend the incubation period and inhibit cough frequency in model mice and guinea pigs(P<0.05), and significantly improve the tracheal phenol red excretion volume as well as serum IL-10 level in mice(P<0.05),besides, the incubation period of asthma extended significantly in guinea pigs(P<0.05). Conclusion: The couplet medicines Fritillaria thunbergii and Rhizoma anemarrhenae has significant effects on relieving cough, eliminating phlegm and relieving asthma, the mechanism of which may be related to peripheral anti-inflammation.
