摘要
为探讨促卵泡素受体(FSHR)基因mRNA在不同组织中的相对表达量和表达规律,探究其调控机制,根据原鸡FSHR基因的编码序列,跨内含子设计1对荧光定量引物,采用qRT-PCR技术检测,研究组织表达谱及其在性腺轴中的表达规律。结果表明:FSHR基因在卵巢和睾丸中高表达,极显著高于其他组织。FSHR基因的表达呈不断变化的趋势,在母鸡卵巢中出壳时最高,持续下降至16周龄时最低, 20周龄稍有回升;FSHR表达量在公鸡睾丸中也于出壳时最高,持续下降至16周龄时表达量最低。在公母鸡的垂体组织中,FSHR基因表达呈小幅上升,母鸡较公鸡上升幅度稍大,卵巢中的表达量略高于睾丸。这一研究为进一步研究FSHR基因的作用机制和表达调控提供了素材。
In order to explore the expression profile and expression pattern of FSHR gene and its possible mechanism of action, a pair of fluorescent quantitative PCR primers of FSHR was design across introns. qRT-PCR was used to detect the tissue expression profiles and expression patterns of FSHR. The results showed that FSHR gene achieved the highest expression in ovarian and testicular which was significantly higher than that in other tissue(P<0.01). The expression of FSHR gene showed a trend of change, and was the highest in the hen's ovary at birth, the lowest at 16 weeks of age, and slightly increased at 20 weeks of age. The expression level of FSHR was also the highest in the testis of male chicken at birth, the lowest at 16 weeks of age. The expression of FSHR gene increased slightly in the pituitary tissue of male and female chickens, and was slightly higher in the females compared with the males, and the ovarian expression level was slightly higher than the testicle. This study investigated the expression profile and expression pattern of FSHR gene in Jinghai Yellow chicken, and provided materials for further study on the mechanism of action and expression regulation of FSHR gene.
